Outline of Research at the Start

細胞内外の情報伝達・物資輸送を担う、脂質二重膜と膜タンパク質が構成する分子集合体がどのように形成され、その機能が決まるかを明らかにすることを目的とする。人工脂質二重膜系と無細胞合成系を用い、生き物に頼らずにイオンチャネルの4量体を形成して①支持脂質二重膜への再構成と原子間力顕微鏡による分子像観察、②無細胞合成系での4量体形成、③電位印加下での分子構造と薬剤応答のその場観察の3つの課題に取り組む。

Outline of Annual Research Achievements

本研究では、人工脂質二重膜系と無細胞タンパク質合成系からなる膜内分子集合体について、その構造・機能およびこれらが働く機構を明らかにすることを目的とした。支持脂質二重膜(SLB)へのイオンチャネル再構成、分子像観察に取り組んできたが、その過程で多価不飽和脂質によって誘起される脂質ドメイン形成など新規現象も発見した。
フォスファチジルコリン(PC)、多価不飽和フォスファチジルエタノールアミン(PE)およびコレステロール(Chol)からなる三成分SLB内に形成され、膜融合サイトとして働く微小ドメインの膜融合効率を評価した。膜融合は、プロテオリポソームとして取得する膜タンパク質を人工脂質二重膜系に再構成する際に必須の過程であり、その機構を知ることはイオンチャネルなどの膜タンパク質研究のスループット向上にも繋がる。それぞれ緑、赤の蛍光標識脂質を添加して調製したPC+PE+Cholベシクル懸濁液を混合し、蛍光スペクトル計測によりFRETの経時変化を取得することで膜融合の経時変化を追跡する実験系を構築した。多価不飽和PEの濃度および不飽和度に依存した膜融合速度を、定量的に評価することができた。
コムギ胚芽抽出物を用いた無細胞合成系では大豆の粗精製脂質であるアゾレクチンが用いられる。天然由来の多成分混合系である脂質を用いて再現性良くSLBを作製するための手法の1として、2023年度はbicelleを用いたSLB形成を行った。短鎖PCとアゾレクチンを混合して調製したbicelleについて、混合比やSLB形成時の条件を検討することで、変性の少ないSLBを作製することに成功した。
無細胞合成系で発現したK+チャネル(hERGチャネル)についてAFMによる分子像観察から四量体形成効率を評価した。また、Na+チャネルのSLBへの再構成とAFM観察にも取り組み、今後の発展が見込まれる有益な結果が得られた。
