Outline of Research at the Start

フラボノイドなどの機能性食品成分の生理活性発現には生体利用性が大きく影響する。日本人の適応進化に関わる遺伝子のALDH2(アルデヒド脱水素酵素2)はアルコールだけでなく、様々なアルデヒド化合物の代謝に寄与することが知られているが、機能性食品成分の生体利用性に与える影響は不明である。そこで本研究では、日本人特有の遺伝的背景であるALDH2変異が機能性食品成分の代謝・生理機能に与える影響を明らかにする。これらの研究を通して、食品機能の分子基盤を得るだけでなく、「日本人特有の代謝特性と食品機能調節」を理解し、機能性食品開発における新たな指針を提案したい。

Outline of Final Research Achievements

The ALDH isozyme-knockout hepatocytes were successfully established. Using these, we demonstrated that ALDH1A1 plays an important role in total cytosolic ALDH activity. 3-Hydroxyphenylacetic acid, a quercetin glycoside enterobacterial catabolite that does not exhibit antioxidant activity, alleviated the acetaldehyde-induced cytotoxicity by modulating total ALDH activity in an AhR-dependent and Nrf2-independent manner. A combination of quercetin glycoside enterobacterial catabolites cooperatively protected the hepatocytes from acetaldehyde by increasing resistance to oxidative stress through a glutathione-dependent increase in ALDH activity. Furthermore, studies with Aldh2 knockout mice showed that decreased ALDH2 activity did not significantly affect the metabolism of quercetin in the small intestine.

Academic Significance and Societal Importance of the Research Achievements

本研究により明らかとなった、ケルセチン配糖体の主要な腸内細菌代謝物である3-ヒドロキシフェニル酢酸や腸内細菌代謝物の組み合わせ処理による肝細胞保護効果は、近年注目度が極めて高いポリフェノールの健康増進作用に関して、新たな科学的根拠を提供するものであり、食品成分のもつ機能性や安全性の科学的理解に大きく貢献することが期待される。また、本研究で明らかとなった腸内細菌代謝物の細胞保護作用に基づいた新規機能性食品やサプリメントの開発に貢献することが期待される。
