Outline of Research at the Start

細胞膜の脂質二重層に発生する張力の作用はすべての膜タンパク質に及んでいる。しかし、微弱な膜張力を制御して機能解析を行うことの難しさから、膜張力作用の具体的影響については現在も明らかになっていない。本研究では独自に開発した人工膜実験法を基に、膜張力と膜タンパク質機能の関連解明に向けた基盤実験技術を確立させる。そして、典型的イオンチャネルの1分子機能解析を通して、膜張力研究への応用可能性を実証する。これまで事実上看過されてきた膜張力作用を、膜タンパク質制御因子として議論の俎上に載せることで、新たな研究領域の開拓を目指す。

Outline of Annual Research Achievements

本研究で開発した膜張力実験プラットフォームとリアルタイム張力解析システムを用い、KcsAカリウムイオンチャネルの膜張力依存特性を単一チャネル電流測定により詳細に検討した。これまでに、脂質二重層の単層（リーフレット）ごとの張力操作実験により、KcsAチャネルは細胞質側リーフレットの張力を感知していることが示唆されている。そこで、より詳細な張力感知の分子機序を明らかにするため、変異体を用いて張力感知部位の検討を行った。変異導入部位として、膜貫通ヘリックスに存在するトリプトファンやチロシンに着目し、これらをアスパラギンまたはロイシンに置換した変異体KcsAを作製した。また、細胞質側リーフレット表面に配置すると考えられているアミノ末端の両親媒性ヘリックス（M0）の関与を検討するため、M0削除体も実験に用いた。前年度に確立させた膜曲率依存的構造変化の蛍光検出法により変異体のスクリーニングを行ったところ、膜貫通ヘリックスの細胞質寄りに位置するトリプトファンの変異体とM0削除体において、張力感知への関与をうかがわせる実験結果が得られた。現在、これら変異体について膜張力制御下で単一チャネル電流測定を行い、性質の変化を解析中である。さらに、当初の研究計画には無かったが、当実験システムの汎用性を実証するためにKcsA以外のチャネルにも応用する試みを始めた。対象としたTRAAKチャネルは張力感受性を持つことが明らかにされているが、その分子機序は明らかになっていない。TRAAKに対してもリーフレットごとの張力制御下で単一チャネル電流測定を行い、その影響を解析中である。

Reason

KcsAチャネルの膜張力感知機構に関する変異体の検討がおおむね予定通りに進んでいる。さらに、当初の計画には無かったが、本研究で開発した張力実験システムをTRAAKチャネルの解析に応用する試みも始め、成果が得られつつある。

Strategy for Future Research Activity

KcsAチャネルの膜張力感知機構に関しては、スクリーニングにより張力感知部位として候補に挙がった変異体の単一チャネル電流解析を推進し、変異導入による張力依存的なチャネル開閉挙動への影響を評価する。また、本研究で開発した張力実験システムの汎用性を実証するため、KcsAだけでなくTRAAKチャネル研究への応用によっても成果を目指す。
