Outline of Research at the Start

パイロトーシスは炎症誘導性の細胞死であり、主に免疫細胞で観察されるがそれ以外の組織でも観察され、炎症性の傷害に広く関与している。パイロトーシスはインターロイキンなどの炎症促進物質を、ガスダーミンと呼ばれるチャンネルを介して細胞外に放出する機構である。ガスダーミンは細胞内で不活性型前駆体として存在し、カスパーゼによって切断されるとN端側の約30kDaのドメイン(p30)が細胞膜へと移行して会合し膜にチャンネルを形成する。アポトーシスからパイロトーシスへと移行して炎症が主じることもあり、様々な疾患に関与した極めて重要な細胞死機構である。本研究では特に代謝との関係に重点を当てて研究する。

Outline of Final Research Achievements

Regulation mechanism of pyroptosis by metabolites was investigated. Firstly, it was shown that pyroptosis was activated by glycolysis in RAW264.7 murine macrophage cells stimulated with lipopolysaccharides. Inactivation of HIF1, hypoxia-inducible factor, through methylglyoxal seems to be involved in this phenomenon. Second, we found that higher glycolysis is accompanied by not only higher GSDMD expression but also lesser GSDME expression.

Academic Significance and Societal Importance of the Research Achievements

炎症は様々な病態で見られる普遍的な所見であるが、その制御機構はあまりよくわかっていない。パイロトーシスは近年発見された炎症促進性の細胞死であるが、その制御機構の詳細は未だ明らかでない。免疫細胞の一種であるマクロファージにおいて、解糖系はパイロトーシスを促進させることを見出した。これは糖尿病における炎症の増悪などとの関連を予想させるものである。また、二つの全く別なパイロトーシス実行因子であるガスダーミンDとEの発現が解糖系により全く逆方向に制御されていることも見出し、ガスダーミン分子の機能分担に新たな知見を加えた。
