Outline of Research at the Start

本研究の目的は、両性電解質高分子化合物の凍結挙動解析による凍結保護機序の解明である。乾燥/凍結耐性生物が持つ特殊な保護タンパク質との共通性である多数のイオン性残基から、凍結濃縮時の細胞脱水コントロールが凍結保護のメカニズムであるとの仮説を立てた。温度可変固体NMRの手法を独自に開発し、凍結時の分子挙動から、細胞に与える浸透圧、脱水作用を考察し真のメカニズムにせまる。機序の解明により、最適分子設計が可能となり、これまで保存困難であった細胞や組織、臓器の保存技術への応用も期待できる。

Outline of Annual Research Achievements

両性電解質高分子による細胞の凍結保護メカニズムを固体NMRにて解明することを目的としてこれまで研究を続けてきた。本年度は、これまで機序の解明を行ってきたカルボキシル化ポリリジンでの結果をより一般化する意味で、他の両性電解質高分子について検討を行った。カルボン酸とアミンによる両性電解質高分子については、ランダムな共重合体と交互共重合体の比較、スルホン酸と4級アミンによる両性電解質高分子、また分子量の違いなど、パラメータをいくつか変更し比較を行った。
その結果、スルホン酸と4級アミンの両性電解質高分子では凍結保護効果が低い結果が得られた。また交互共重合の方が高い保護性能を得られることがわかった。これらの高分子溶液の低温でのNMR挙動を詳細に調べたところ、凍結保護活性の低いポリマーではバルクの水の凍結温度が高く、凍結時の急激な浸透圧上昇による障害が示唆される結果であった。また、凍結保護効果に関連していると思われるパラメータとして、低温でのナトリウムイオンの運動性が挙げられる。低温における水の運動性低下に伴いナトリウムイオンがポリマー-水マトリックスにトラップされることで運動性が低下し、浸透圧への寄与が低下することで細胞脱水を適度に制御している可能性がある。分子量や高分子の構造により、水やナトリウムイオンの取り込みの度合いが変化することで凍結保護活性にも違いが見られたと考えられる。このように、凍結過程における様々な溶液のダイナミクスパラメータを固体NMRで測定することにより、細胞凍結保存に重要な考察を与えたことは特筆に値する成果と考えられる。論文発表を準備中である。
