Outline of Research at the Start

生命の起源から現在に至るまで、RNAやRNP（RNA-タンパク質複合体）は生命機能の制御に重要な役割を果たしている。しかし、その解明はいまだ十分ではない。本研究では、細胞の機能制御を担うRNAやRNPに注目し、細胞内での制御ネットワークや作動原理を解明する。さらに、その理解に基づいて細胞の機能を制御する人工RNA/RNPシステムを構築し、医療や生命工学上有用な技術を開発する。また、RNA/RNPに基づく生命の起源モデルを構築し、生命システムの創発原理の解明を目指す。以上、RNAやRNPが司る生命現象の理解と制御を通じて、「合成生命システム創生分野」を切り拓く。

Outline of Annual Research Achievements

RNAを基盤とする生命システムが細胞の機能制御に果たす役割の多くは未知のままである。またRNAやRNA-Protein (RNP)相互作用は、生命の進化の過程で本質的な役割を果たしたと考えられるが、それらがいかに細胞や生命のシステムを形成するに至ったのか、その構築原理の解明には至っていない。本研究では、(1)細胞機能を制御するRNP相互作用ネットワークを包括的に同定するとともに、(2)RNPによる細胞内構造物の作動原理を解明し、細胞内で機能する人工RNPを構築する。また、得られた知見を生かして、(3)哺乳類細胞や個体で作用する人工のRNA・RNPシステムを開発する。同時に、(4)生命システムの創発原理の解明を目指し、人工RNA細胞モデルの創出に挑む。
本年度は主に、(3)人工のRNA・RNPシステムの基盤技術の開発に取り組み、マイクロRNA（miRNA）に応答して遺伝子発現を活性化させる合成mRNA（miRNA応答ONスイッチ）を開発することに成功した。このシステムはmiRNAを発現している細胞でだけ遺伝子を発現させることができる。これにより、iPS細胞やiPS細胞から分化した心筋細胞の高効率での純化が可能となった。加えて、光やタンパク質に応答して遺伝子発現を活性化させる仕組みも開発した。今後、これらの様々な翻訳制御システムの実装による、機能性人工細胞の構築に挑戦する。

Reason

本年度は主に、(3)人工のRNA・RNPシステムの基盤技術の開発に取り組みんだ。その結果、マイクロRNA（miRNA）に応答して遺伝子発現を活性化させる合成mRNA（miRNA応答ONスイッチ）を開発した。このシステムはmiRNAを発現している細胞でだけ遺伝子を発現させることができる新規なシステムである。さらに、光やタンパク質に応答して遺伝子発現を活性化させる仕組みも開発した。このように、今後の機能性人工細胞の構築において有用なツールとなりうる基盤技術の開発に成功した。また、(2)RNPによる細胞内構造物の構築においても、細胞内における人工RNP顆粒の形成を示唆するデータなどが得られていることから、おおむね順調に進展していると判断した。

Strategy for Future Research Activity

【RNP構築原理の理解に基づく人工オルガネラの設計と構築】
細胞内における人工RNP顆粒の形成を示唆するデータが得られため、より詳細な解析を行い、人工的にデザインしたRNAおよびRNA結合タンパク質を使った顆粒の形成原理の詳細についての理解を得る。
【医療応用に資する機能性人工RNA/RNPシステム・細胞の開発】
これまでに構築したmiRNA応答スイッチを中心とした人工mRNA技術を統合し、RNAを基盤とする人工遺伝子回路の構築と細胞制御技術を開発する。また、マウス個体への適用を進める。
【生命進化における人工RNA/RNP 細胞モデルの創成】
RNAからなる小胞構造の形成について解析を行い、RNA細胞モデルの設計原理の確立を目指す。
