Outline of Research at the Start

植物ミトコンドリア (mt) ゲノムはエネルギー生産に必須の遺伝子や重要農業形質 の原因遺伝子等をコードしているため, 基礎科学的にも農業生産的にも重要な研究と改変の対象であるにもかかわらず, これまでゲノム改変技術が不在であったため, 手つかずの最後のゲノムとしてとり残されていた. 本研究では, 我々のグループが近年開発した植物mtゲノム改変技術を活用し, mtゲノム遺伝の基礎的性質を明らかにし, またmtゲノム改変集団を通じて育種応用の潜在性検証と重要農業形質に関与する遺伝子の同定を行い, 世界に先駆けてmtゲノム育種基盤の開拓を目指す.

Outline of Final Research Achievements

In this study, we developed organelle genome editing technologies for plant mitochondria and chloroplasts, enabling precise gene knockouts and base editing. By applying mitoTALEN and TALECD technologies, we successfully identified and disrupted cytoplasmic male sterility (CMS)-associated genes such as orf352 in rice and orf218 in eggplant. Furthermore, chloroplast genome base editing allowed the introduction of agronomically beneficial traits such as herbicide resistance. These advances provide a new foundation for crop improvement and plant reproductive biology.

Academic Significance and Societal Importance of the Research Achievements

本研究は、植物ミトコンドリアおよび葉緑体といったオルガネラのゲノム編集を高精度に実現し、育種上重要な細胞質雄性不稔性の分子機構解明や原因遺伝子の直接改変技術を確立した点で学術的に画期的である。また、葉緑体ゲノムの精密な一塩基改変による除草剤耐性導入にも成功し、「オルガネラゲノム編集育種」という新しい流れを作り出した。これらの成果は、F1品種安定生産や効率的な品種改良実現、農薬使用量と環境負荷低減といった持続可能な農業への貢献が期待される。植物オルガネラゲノム編集技術は、他の種や遺伝子にも応用可能であり、基礎研究から応用展開まで幅広い波及効果と、国際的にも高い学術的・社会的意義を有する。
