Outline of Research at the Start

植物の表皮に存在する気孔は、光合成に必要な二酸化炭素の唯一の取り入れ口であり、変動する環境条件に応答してその開度を調節している。これまでの研究により、気孔を構成する孔辺細胞では、光に応答して細胞膜プロトンポンプが活性化され、気孔開口の駆動力を形成することなど、気孔開・閉のシグナル伝達の一端が明らかとなってきたが、詳細な分子機構は未だ不明の部分が多い。本研究では、生理・生化学・遺伝学的手法やケミカルバイオロジーを駆使し、気孔開・閉のシグナル伝達の分子機構を明らかにする。さらに、これらの成果に基づき気孔開度を人為的に制御する技術開発を行い、植物の成長促進や乾燥耐性の付与の技術確立を目指す。

Outline of Annual Research Achievements

気孔開口の必須酵素である細胞膜H+-ATPaseは、C末端から2番目のスレオニンのリン酸化により活性化されることが明らかとなっているが、リン酸化制御に関わるキナーゼやホスファターゼは明らかとなっていない。これまでの生化学的な解析により、タイプ2Cホスファターゼ(PP2C)様の活性が脱リン酸化に関わることがわかっていたため、PP2Cの分子種の同定を進めた結果、PP2C.Dファミリーが重複して機能しており、その中でも孔辺細胞での発現量の多いPP2C.D6とPP2C.D9が主要な役割を果たしていることが明らかとなった。この研究過程において、暗処理した植物では急激に細胞膜H+-ATPaseが脱リン酸化されることを見出し、PP2C.D6とPP2C.D9が葉内CO2の急激な上昇に伴って活性化され、細胞膜H+-ATPaseの脱リン酸化に関わっていることが明らかとなった。
また、栄養生長期と生殖生長期に出現する葉における気孔特性の解析を進め、生殖生長期の葉の気孔は、栄養生長期のものより光による気孔開口が促進されており、光合成活性が高く、種子生産に重要な役割を果たしていることが明らかとなった。さらに、気孔開度に影響を与える化合物の網羅的探索の結果、植物由来の天然物が強い気孔開口抑制作用を持つことがわかり、その分子機構についての解析を進めた。
加えて、気孔開度制御による植物の成長や収量に与える影響についての解析も進め、ゲノム編集など遺伝子組換えに頼らない方法による「ポンプ植物」の作出を目指した解析を進め、シロイヌナズナの主要なH+-ATPaseアイソフォームのプロモーター領域にガイドRNAを設計し、ゲノム編集により変異を導入することで、発現量の増加した植物体の作出に成功した。表現型解析の結果、これらの植物では、気孔開口が促進されており、バイオマスが25%以上増加していた。

Reason

PP2C.Dの機能解析やCO2による活性制御など気孔の光シグナル伝達の分子機構の解析が大きく進展した。また、植物由来の天然物が強い気孔開口抑制作用を持つことがわかり、その分子機構の解析も進展した。加えて、プロモーターをゲノム編集により改変することで、標的遺伝子の発現量の制御にも成功した。

Strategy for Future Research Activity

細胞膜H+-ATPaseとPP2C.Dを基軸として、光シグナル伝達の上流因子の探索や機能解析をさらに進める。また、ケミカルスクリーニングにより得られている多数の気孔開度に影響を与える化合物の分子機序の解明を進める。加えて、プロモーターをゲノム編集により改変することで標的遺伝子の発現量の制御した植物について詳細な表現型解析を進めるとともに、細胞膜H+-ATPaseのネガティブレギュレーターの変異体は、気孔開口が促進されていることがわかってきたので、それらの詳細な表現型解析も進める。
