Outline of Research at the Start

本課題では,プログラム通りに自動的に活動し物質生産する人工細胞群を「オートマター」と呼び,その実現を目指す.そこで不可欠となる動作制御のために,分子制御,電子制御とそのインターフェイスを開発し,人間のプログラムに従うよう設計を行う.分子分解能での設計からマクロな仕事を実現するために,大量の人工細胞を群れとして協調動作させるためのメカニズムと原理を探求する.

Outline of Annual Research Achievements

本研究では,プログラム可能な自動微粒子群・オートマターの実現に向けた要素技術の開発と統合を目的とする。 本年度も,課題の要素技術として4つのサブテーマ1)自己複製能,2)分子制御,3)自動大量生産,4)電子制御-分子インタフェイスの実現と,これらの統合を目指した研究を進めた。1)について,清水グループでは,前年度までに見出した2種類に加えてさらに1種類のタンパク質因子を発見し,活性のあるリボソームの自己組織化ルートを見出した。これは,PURE自己複製溶液の実現に向けた重大なステップと認められる。2)について,大野グループでは,オートマター内部における自律的かつ選択的な遺伝子発現制御系の実現に向けて,特定の分子の存在を検知して任意の遺伝子の発現を誘導できる新規なシステムを開発し報告した。これは,転写翻訳系を制御するための基盤技術としての活用が期待できる。3)について,野村グループでは,オートマターの筐体となる人工多細胞体を簡便かつ大量に生産する手法の研究を推し進め,ロボットを用いた生産と評価の自動化にも成功し報告した。そのcmスケールの立体構造を用いた内外溶液の安定性評価を行い,核酸情報のセンシングおよび内包させた薬剤の徐放系としての動作検証を進めている。4)について,瀧ノ上グループでは,オートマターの自律的な運動を実現するために,マルチブランチ型RNAモチーフを自己組織化させてRNA液滴を生成する技術を開発し,分子設計に基づく安定性の制御方法を得た.そしてこれらサブテーマの統合として,人工設計核酸によるマイクロサイズ構造の制御手法についての知見が得られている。以上の成果は,分子分解能で設計された分子システムが,よりマクロなスケールの動作を制御可能であることの実証例であり,オートマター実現とその利用に向けた重要な進歩である。

Current Status of Research Progress

1: Research has progressed more than it was originally planned.

Reason

2023年度は要素技術開発のうち,複数の項目で当初の計画以上の進展がみられた。1)について,本課題で最も困難と予想している自己複製機構の実現に向けたリボゾーム再構成実験について重要な進展があった.いまだ達成されていない転写rRNAからのリボソーム大サブユニットの再構成についての検討を行っている。転写rRNAからの再構成に加え、いくつかのrRNA塩基修飾酵素を含む形での再構成はともに活性のある大サブユニットの検出には至らなかった。一方、個々の塩基修飾酵素欠損株ライブラリー(東京大学・市橋教授より提供)を用い、調製したrRNAを用いた大サブユニットの再構成を行ったところ、新たなGTP加水分解酵素依存的に再構成が促進されることを見出した。すなわち、これまでに明らかにした2つの酵素に加え、さらに1つの因子が活性のある大サブユニットのアセンブリに大きく関与することが新しく明らかになった。これは転写翻訳に基づくオートマターのプログラム可能性において極めて重要な意味を持つ成果である。2)について,オートマター内部または生体内部において特定の分子に応答して遺伝子発現を制御することを可能にする標的依存性RNAポリメラーゼTdRNAPの開発に成功し,Nature Comm.誌に報告した.融合抗体を置換することにより,ペプチド、タンパク質、RNA、小分子などのさまざまな分子に応答して目的の転写物を生成することが実証された.また4)について,これまでの光応答DNA液滴に加えて人工設計RNA液滴を生成する技術を開発し,安定性をコントロールする方法を得て論文投稿中である。さらにこの液滴にmiRNA認識部位を導入し,miRNA有無のAND演算を行って検出することにも成功した。これらは研究開始当初に予想しなかった重要な結果であり,総じて本年度は当初の計画以上に進展したと考えている。

Strategy for Future Research Activity

本課題の要素技術として4つのサブテーマ1)自己複製能,2)分子制御,3)自動大量生産,4)電子制御-分子インタフェイスの実現と,その統合によってオートマターの具現化を目指す。その実証を通じて,物質科学と生命現象との融合分野を情報化して制御する新たな科学的基盤に挑む.最終年度となる本年度は4つのサブテーマおよびその成果の統合を目指す。サブテーマ1)引き続き,人工的に調製した構成要素を用いたリボソームの自己組織化に注力し,無細胞PURE自己複製溶液の構築を目指す。また他のサブテーマとの連携による人工細胞への組み込み研究を進め,オートマターとしての自動物質生産系の実装を目指す。サブテーマ2)これまでに構築したRNP分子制御システムを発展させ,人工細胞内部からの転写・翻訳制御手法の検討を行う。特に,本課題で見出した新たな無細胞RNP制御系の研究を進め,自律的かつ選択的な遺伝子発現の制御手法の確立を目指す。サブテーマ3)人工多細胞体のコンパートメント間相互作用/内外情報伝達システムの実現に注力する。複数種のコンパートメントを共存させた系で,外部からの核酸情報の取り込みとコンパートメント間通信等の検証を行う。サブテーマ4)外部制御可能な人工細胞分子デバイスとして,光応答性DNA液滴・RNA液滴をオートマターの処理装置として利用するための開発に取り組む。これらの各要素技術とその連携動作について,分子による内的制御と外的な制御とに注力した機能の具現化と評価・報告を進める。
