Outline of Research at the Start

本研究は超音波による細胞機能の制御技術を応用することで,生体外における立体心筋組織への血管新生の促進手法を具現化するものである.このために下記の3ステップを超音波を用いることで遂行する.①培養平面上で血管新生に適した初期位置に血管内皮細胞を配置し,心筋細胞との共培養を行う.②血管内皮細胞と心筋細胞を共培養した細胞群をシート状に剥離する.③こうして剥離した細胞シートを積層し,適切な出力の超音波照射により立体組織の血管新生を促進する.

Outline of Annual Research Achievements

本研究は再生医療における課題の一つである立体細胞組織の活性を維持し,生体外細胞組織を作製するための基盤技術・基盤知識を構築することを最終目標として実施した.具体的には超音波の照射により血管新生を促進することを目的として研究を始めた.実際に血管内皮細胞(Human Umbilical Vein Endothelial Cells:HUVEC)への超音波照射が血管新生を促進する可能性について原著論文にまとめた.具体的な内容を記す.形態観察により,コラーゲンゲル内に置いて3次元培養された血管内皮細胞に対して超音波を照射することで,細胞が管腔構造を構築すること,ネットワークを構築することを確認した.さらに,タンパク質の発現を確認することで,血管新生に関するホルモンの発現が超音波によって上昇することを証明した.その一方で,その原理は未だ不明確であったため,原理解明のため複数の研究を並行して遂行した.
超音波をはじめとした機械的な刺激をコントロール可能な細胞培養装置を開発し,立体組織の培養を行うためのプロトコル構築を行なった.具体的には,金属製の3Dプリンタを用いて機械刺激をコントロールできる培養装置を開発した.この結果をまとめた論文が本年度一報受理されている.さらに,超音波照射が培養液の流動を発生させて,その流動が細胞の活動を制御するという内容の原著論文も一報受理されている.また,培養液の灌流が3次元細胞組織の活性を向上させるという内容の論文も公表した.さらに,超音波によって細胞の遊走を制御できることも明らかにし,この成果でも論文を執筆した.このように,超音波によって血管内皮細胞の血管新生を促進できるということについて,現象論にとどまらず,原理の解明に向けた研究を複数並行して遂行した.
