Outline of Research at the Start

社会性昆虫は可塑性のパターン（反応規準）をグループサイズに依存して変化させると考えられるが，その仕組みは不明である．グループサイズが可塑性のパターンに与える影響を調べるには，個体レベルにとどまらず，グループサイズの変化に伴う個体間相互作用の構造変化という上位階層を対象にした研究が必要である．そこで，ネバダオオシロアリの初期巣を材料に，女王によるグループサイズの認識機構およびそれに依存した繁殖形質の制御機構を明らかにする．これを解明する上で，本研究課題ではグループレベルから生理・遺伝子レベルに至るまでの階層を分野横断的に整理する．

Outline of Annual Research Achievements

本研究課題は真社会性昆虫であるシロアリの初期巣を対象に，雌雄生殖虫の表現型が巣内の個体数増加に伴って可塑的に変化する可能性を探った．2021年度までに，系統的に祖先的なグループに属するネバダオオシロアリにおける雌生殖虫の産卵は一年に一度きりでありことが示された．また，初期巣における個体間相互作用に応じた行動パターンの変化を解析し，雌雄生殖虫の加齢に伴った繁殖と防衛の巧みな切り替えが生じる可能性を明らかにした．しかし，共通したスイッチで繁殖と防衛が切り替わる可能性は低く，繁殖能力を低下させる要因は攻撃性とは別である可能性がある．そこで最終年度は，営巣後の時系列に伴った生殖虫における繁殖関連遺伝子の発現解析に注力した．生殖虫における繁殖関連遺伝子の発現解析に取り組んだ．2014年にネバダオオシロアリのゲノムが解読され，当論文にて繁殖関連遺伝子がいくつか報告されていた．そこで，初期巣の営巣直後，50日目，100日目における雌雄生殖虫を回収した．対象遺伝子は，女王において発現が確認されているビテロジェニン遺伝子（Yaguchi et al. 2022）および雄の精子形成遺伝子を対象にプライマーを設計した．これらの遺伝子について，リアルタイム定量 PCR 法による遺伝子発現解析を実施した．精子形成遺伝子の発現量は，個体間のばらつきが大きいものの，営巣後の時系列に沿って変動することが示唆された．一方で，ビテロジェニン遺伝子は発現量のばらつきが大きく，特徴的なパターンを得ることができなかった．生殖虫の解析個体数は限られており，いずれの遺伝子においても更なる検証を要すると考えている．
