Outline of Research at the Start

矯正学的歯の移動の際に発現するTNF-αは、破骨細胞形成に重要な役割を果たしている。近年、細胞死の研究の進展により、プログラムされたネクローシスとして新しい概念のTNF-αによるネクロプトーシスが脚光を浴びている。矯正学的歯の移動時の圧迫側では、歯槽骨内にある骨細胞は細胞死を起こしていくことが報告されている。本研究は、矯正学的歯の移動時のTNF-αによる骨細胞のネクロプトーシスと、その際に放出されるIL-33の破骨細胞形成への影響を調べる研究である。

Outline of Annual Research Achievements

矯正学的歯の移動において骨細胞が細胞死するときのメカニズムは不明な点が多い。また、矯正学的歯の移動時における骨細胞のネクロプトーシスとdamage-associated molecular patterns (DAMPs)による破骨細胞形成との関連性について明らかにされていないのが現状である。さらに、DAMPsのひとつであるIL-33によるTNF-α依存性の破骨細胞形成について調べた報告はまだない。本研究では、矯正学的歯の移動時のTNF-αによる骨細胞のネクロプトーシスと、その際に放出されるIL-33の骨代謝への関連性を調べることを目的として研究を行った。
まずは、in vitroとin vivoでTNF-αによる破骨細胞形成と骨吸収にIL-33が与える影響について調べた。IL-33はTNF-α依存性の破骨細胞形成および骨吸収能を有意に阻害することが確認できた。また、Western blottingと免疫蛍光染色により、IL-33はIκBのリン酸化とNF-κBの核内移行を抑制することで、破骨細胞形成を阻害することが示唆された。次に、矯正学的歯の移動のマウスモデルで12日間歯を移動させた時のIL-33の影響を調べた。IL-33を投与した群は、IL-33を投与しなかった群と比較して歯の移動量と破骨細胞形成が有意に減少した。したがって、IL-33は圧迫側における破骨細胞形成を阻害することで矯正学的歯の移動を抑制することが示唆された。さらに、ネクロプトーシスの指標となるp-RIP3を矯正学的歯の移動時の骨細胞から検出した。以上より、矯正学的歯の移動時の骨細胞のネクロプトーシスが破骨細胞形成に影響を与えていると推測される。
