Outline of Research at the Start

プリオン病原体は現代医療では治療不可能な神経変性疾患「プリオン病」を引き起こす。本研究では、プラズマ活性溶液がプリオン感染神経細胞に対して選択的に細胞死を誘導できるかを明らかにするとともに、プラズマ活性溶液がプリオン感染神経細胞に誘導する細胞死の分子機構を解明する。プリオン感染細胞がどのようにして除去されていくのかが明らかになることで、治療応用への展開が期待できる。

Outline of Annual Research Achievements

2021年度(令和3年度)から2022年度(令和4年度)にかけて、さまざまな気体由来のプラズマで調整したプラズマ活性溶液のプリオン感染細胞に対する細胞死誘導効果について解析した。LDH(乳酸脱水素酵素)放出アッセイで死細胞の割合を比較したところ、 N2 >CO2 >O2 >Ar >Airの順番でプリオン感染細胞(ScN2a)と非感染細胞 (N2a)の死細胞の割合の差が大きかった。さらに、ELISA(Enzyme-linked immunosorbent assay)を用いたモノおよびオリゴヌクレオソーム定量によるアポトーシス(Apoptosis)の解析を同様に行ったところ、N2 >O2 >CO2 >Air >Arの順番でアポトーシス量の差が大きかった。これらのことから、由来する気体によりプラズマ活性溶液に含まれる活性種が異なることでApoptosisなどの細胞死を引き起こす効果に違いが見られたものと考えられた。
このため、2023年度(令和5年度)は活性種を消去するラジカルスカベンジャーを空気由来のプラズマ活性溶液に対して用いて、ScN2aとN2aの細胞死への活性種の影響の解析を行った。活性種の消去剤として、・OHに対してはDMSO、ONOO-に対してはUric acid、・NOに対してはCarboxy-PTIO、H2O2に対してはCatalase、・O2-に対してはSODを用いた。その結果、消去剤を加えていないコントロールと比べて、N2aでは・O2-を消去時に細胞傷害性が有意に低下しており 、一方ScN2aではONOO-と・O2-を消去時に細胞傷害性が有意に低下していた。
