Outline of Research at the Start

本申請研究では食品からのボツリヌス菌検査法において、国内の様々な検査室で技能試験に利用可能な無毒化株の開発を目指す。すなわち、(1)毒素遺伝子をノックアウトした無毒化株を作成し、(2)親株と無毒化株を利用した食品への添加・回収試験を比較検討し無毒化株の標準株としての妥当性確認を行う。

Outline of Final Research Achievements

In addition to development of standard method, construction of procedure for quality control is required for internationally acceptable microorganism testing system. In this study, we developed standard procedure for preparation of inoculum which can be used in recovery test for standard method of Clostridium botulinum detection. In addition, we assessed LOD50 for the standard method which required in implementation verification described in ISO 16140-3. This information enables internationally acceptable quality verification of testing results produced in laboratories they use domestic standard method for Clostridium botulinum detection.

Academic Significance and Societal Importance of the Research Achievements

食品衛生検査およびその精度管理に関する国際的整合性の確保は、食品流通のグローバル化等に対応するために、本邦の食品衛生行政において大きな課題となっている。本研究で得られた成果は、ボツリヌス菌標準試験法に対する国際基準を満たした技能試験の国内での実施を可能とするのみならず、法的制限により取り扱いが困難な菌種に対する技能試験実施方法のモデルケースとなるものであり、本邦の食品衛生行政の国際化対応に寄与するものである。
