Outline of Research at the Start

エドワジエラ症は Edwardsiella 属細菌による感染症の総称で、特にヒラメ、マダイ、ウナギの養殖における魚病被害では一番の問題となっている。本菌にはIII 型分泌装置と呼ばれる針のようなタンパク質輸送装置が存在し、前申請研究でIII 型分泌装置から分泌される2つの遺伝子(orf13及びorf19)がヒラメの病原性に関与することを明らかにした。本研究では両遺伝子に着目し、分子生物学的な解析を進めることにより、III 型分泌装置を介した本菌の未知の病原性発現機構を明らかにする。一方で両遺伝子ノックアウト株を生菌ワクチンとして利用することで本菌による魚類エドワジエラ症の予防法の開発を目指す。

Outline of Final Research Achievements

In previous JSPS KAKENHI (16K07849), the type III secretion system (T3SS) was associated with Edwardsiella piscicida virulence. The virulence of E. piscicida mutants carrying insertion mutations in orf13 (EscE) and orf19 (EsaE) was significantly reduced in Japanese flounder, whereas that of E. piscicida mutants carrying insertion mutations in orf29/30 (EseJ) was not affected. Intracellular replication activity in macrophage-like cells isolated from Japanese flounder was diminished in all four mutants. These results were recently published. However, the vaccine effects of orf13 and orf19 mutants were not observed.
Furthermore, T3SS-translocon protein expression was reduced when bacterial cells were cultured at high NaCl concentrations; however, epithelial cell adherence increased at high NaCl concentrations. These results suggest that E. piscicida first adheres to fish epithelial cells by downregulating T3SS expression in marine environments.

Academic Significance and Societal Importance of the Research Achievements

本研究成果から、われわれは本菌によるヒラメエドワジエラ症の新規感染症予防法創製のシードとなる標的遺伝子として、本菌のIII型分泌装置の遺伝子クラスター内に存在するorf13及びorf19遺伝子を同定することに成功した。しかしながら、orf13及びorf19遺伝子ノックアウト株をヒラメに対するワクチン候補株として応用する研究を行ったが、延命効果に止まり顕著なへい死抑制効果は認められなかった。
一方、本菌の宿主への病原性発現機構の初期ステップとして、海水環境ではIII型分泌装置の発現を抑制しつつ、一方で付着能を高めることで先ずは宿主細胞に付着することを優先している可能性が推察された。
