Outline of Research at the Start

本研究はナノ粒子を獣医療分野に応用しようとするものであり、犬や猫に対する高精度放射線増感療法の基盤研究になることが期待される。また獣医療領域では、シスプラチンやゲム
シタビンを放射線増感剤に用いた放射線治療が報告されているが、必ずしも良い成績が得られている訳ではなく、そのため放射線増感剤を用いた報告はほとんど認められない。獣医学分野でのナノ粒子を用いた放射線増感剤を実症例に投与した報告は、申請者らの知る限りでは報告されていない。申請者らは先行研究の中で過酸化チタンナノ粒子が金ナノ粒子を上回る放射線増感効果を発揮することを動物実験において確認し、世界的に高い評価を得ている。

Outline of Annual Research Achievements

申請者らのグループではこれまでに医工連携共同研究の中で、多数の無機物ナノ粒子、有機化合物ライブラリーの中から、ROSの生成量を飛躍的に増加させる物質を網羅的に探索し、二酸化チタンを原材料にした過酸化チタンを独自の手法で、約50-70ナノメートル(nm)にナノ粒子化することに成功し、過酸化チタンナノ粒子では、X線照射により数十倍以上の大量のヒドロキシラジカルを生成することを初めて発見し、細胞内に確実に取り込まれ、X 線照射によって大量のヒドロキシラジカルを発生することを発見した。このROSの産生という特徴は、現状の二酸化チタンでは認められないがん治療において有利な効果である。この過酸化チタンナノ粒子は、良好に腫瘍細胞にエンドサイトーシスで取り込まれることなどを発見し、申請者らが開発した過酸化チタンナノ粒子は生物学的国際調査を行った上で新たな放射線増感剤として米国、欧州、中国等、世界5か国以上で国際特許を得た。この過酸化チタンナノ粒子に関しては、マウス悪性黒色腫などの腫瘍モデルを用いた検討では、免疫療法との併用により、極めて優れた腫瘍増殖抑制効果を発見するなど複数の研究成果を得ている。しかしながら、コロナウイルス感染症などの流布により獣医学における実験動物の作成や実際に大型実験動物腫瘍モデルへの放射線治療においてリニアックを用いて治療計画をした上で、IMRTの適用やその比較に関しては実施することが困難であった。イヌネコなどの腫瘍に関しては、肉腫などの放射線抵抗性腫瘍が多くの割合を占め、放射線抵抗性の克服は重要な課題と考えられるため、過酸化チタンナノ粒子の放射線増感作用を更に調査した上で、一回線量を増やした単回照射や寡分割照射などの放射線の照射方法を再検討することにより、獣医学にとってより最適な治療方法を開発していく予定である。

Reason

獣医学において疾患動物に対してリニアックを用いて放射線治療を実施、検討できる施設は極めて限定されるが、コロナウイルス感染症などの流布により、研究者の移動が困難になり、獣医学における実験動物の作成や実際に大型実験動物腫瘍モデルへの放射線治療においてリニアックを用いて治療計画をした上で、IMRTの適用やその比較に関しては実施することが困難であった。今後は体制を再構築し、再検討していく予定である。

Strategy for Future Research Activity

獣医学における実験動物へのIMRTなどを用いた放射線治療方法の検討は本研究機関内では困難であったが、自施設で行った小動物などを用いた検討では、異なる観点からの抗腫瘍効果を確認できている。免疫チェックポイント阻害剤などの併用をすると、過酸化チタンナノ粒子、放射線療法、免疫療法の併用で優れた治療効果が得られえる予備試験結果を得ており、今後は体制を再構築し、新たな知見を加えつつ、再度研究体制を再検討していく予定である。
