| Outline of Research at the Start |
視床を含む大脳基底核は、運動、認知、情動などの様々な脳機能を調節し、その異常は種々の精神・神経疾患の病態と深い関連を持つことが知られているが、神経回路レベルでの機能的な解析は進んでいない。本研究では、認知機能障害を伴うパーキンソン病において視床の神経細胞が一部変性することに着目し、視床線条体経路の認知機能における役割を解析することで、大脳基底核ネットワークの機能マッピングに取り組む。

| Outline of Final Research Achievements |
The thalamus, a relay point for sensory information, is thought to control motor and learning behaviors by providing input to the striatum, a central structure of the basal ganglia that controls motor control and cognitive functions. In this study, we demonstrated that input from the parafascicular nucleus of the thalamus to the dorsolateral anterior striatum plays an important role in learning functions. In addition, we have successfully developed a new system vector that enables the control of sophisticated gene expression switching to carry out this study. This system is expected to provide a highly effective tool for various aspects of life science research and to be used for developmental purposes in both clinical and basic medicine.

| Academic Significance and Societal Importance of the Research Achievements |
これまで十分な機能解析が進んでいなかった視床束傍核の神経細胞は、背外側前方線条体への連絡によって学習を介した行動制御を担うことを明らかにし、認知機能の獲得に極めて重要なネットワークを形成していることを発見した。 パーキンソン病患者の死後脳剖検から、束傍核を含む視床髄板内核を構成する神経細胞が欠落することが知られている。今回の研究によって、視床髄板内核の認知機能への関与が明らかとなり、このことはパーキンソン病などの疾患の病態解明に繋がるとともに、この経路を制御する薬剤を探索するなど、病態の改善に繋がる薬剤の開発への可能性が期待される。
