Outline of Research at the Start

自閉症スペクトラム障害(ASD)では,その臨床的異質性と多様な併存疾患から,個々の臨床特性を踏まえた客観的評価法の開発が急務とされている.一方,神経ネットワーク障害はASDの神経基盤の有力な候補であり,また神経活動の時間的変遷である“神経ネットワークダイナミクス”はASDの病態生理を理解する上で重要な鍵を握る.
本研究では,申請者らが開発した動的位相変動解析を脳磁図に適用することで神経ネットワークをダイナミカルに捉え,ASDにおける病態生理の新たな一面を浮き彫りにする.さらにクラスタリング分析や脳活動シミュレータを用いて臨床的異質性や併存する精神疾患に配慮した個別化治療システムの構築を目指す.

Outline of Final Research Achievements

Due to its clinical heterogeneity and comorbidity, clinical evaluation based on individual clinical characteristics may improve diagnostic accuracy. On the other hand, aberrant neural network hypothesis reportedly plays a crucial role in neural basis of autism spectrum disorder (ASD). This study aimed to explore a new aspect of network dysfunction of ASD using a dynamical phase synchronization approach applied to MEG. We additionally applied clustering analysis and brain activity simulator aiming to establish a personalized treatment system.

Academic Significance and Societal Importance of the Research Achievements

脳活動シミュレータにおいて,脳生理学的に観測される発火パターンを再現する神経システムを構築し,興奮性/抑制性ニューロン比率や局所的ネットワーク構造,領野間結合に関するパラメータに着目してASD特徴を生成するダイナミカル神経ネットワーク構造の病理パラメータ領域の探索を検討した(Nobukawa et al., 2021).また,カオス共鳴機構を利用し,注意欠如多動症の乱れを即応的に正常化するアルゴリズムを開発し,モデルシミュレーションでの効果検証を実施した(Nobukawa et al., 2021).これらの手法の開発は個別化治療を講じる上で重要な役割を果たすと考えられる.
