| Outline of Research at the Start |
腹部大動脈瘤は、動脈壁の局所的脆弱性をもとに進行性の動脈径の拡大を特徴とする予後不良疾患である。現在、治療法として外科的介入があるが適応が限られるため、薬物療法の開発が望まれている。しかし、腹部大動脈瘤形成機序の詳細が不明のため、有効な治療薬は開発されていない。C-X3-C chemokine receptor 1(CX3CR1)は、ケモカインの一種であるフラクタルカインの受容体である。本研究の目的は、分子生物学的手法を用いてCX3CR1の腹部大動脈瘤形成機序への関与を明らかにし、新規分子標的療法の礎とすることである。

| Outline of Final Research Achievements |
Peripheral blood monocytes are not heterogeneous but are divided into two subsets: inflammatory and non-inflammatory monocytes. We have demonstrated that an increase in non-inflammatory monocytes is involved in the development of future coronary artery disease. On the other hand, both inflammatory monocytes and interleukin 34 are elevated in acute myocardial infarction, suggesting that they are involved in myocardial remodeling and the development of chronic cardiovascular events.Non-inflammatory monocytes are elevated in abdominal aortic aneurysms, and inflammatory abdominal aortic aneurysm formation is suppressed in fractalkine knockout mice, suggesting that fractalkine receptors and non-inflammatory monocytes may contribute to the aortic aneurysm formation process.

| Academic Significance and Societal Importance of the Research Achievements |
ヒト末梢血単球サブセットの評価が腹部大動脈瘤におけるリスク層別化に寄与することが示唆された。また単球サブセットやインターロイキン34を含む炎症性サイトカインの上昇は冠動脈疾患や心房細動発症への関与も示唆されるため、さらなる機序検討が必要であるが、新規治療の礎になると考えられた。
