Outline of Research at the Start

最近のゲノムワイド関連研究で、アポA-Vの変異は心筋梗塞のリスクとして注目されている。アポA-Vの主な生理機能はリポ蛋白中の中性脂肪の異化に関わると考えられている。しかし、動脈硬化発症・進展におけるアポA-Vの病理学的意義はまだ十分に解明されていない。本研究ではリポ蛋白代謝がヒトに類似したウサギモデルを用いて、アポA-V欠損ウサギを作製し、アポA-Vの動脈硬化発症・進展と血中中性脂肪代謝における役割について明らかにする。

Outline of Final Research Achievements

Apo A-V knockout (KO) rabbits were generated. Apo A-V KO rabbits showed increased plasma triglycerides (TG) and TG-rich lipoproteins levels. The elevations were exacerbated by a high cholesterol diet and postprandial state, suggesting a relationship between apo A-V and postprandial hyperlipidemia. Apo A-V deficient rabbits may become a valuable model for studying human triglycerides metabolism, postprandial hyperlipidemia, and atherosclerotic diseases.

Academic Significance and Societal Importance of the Research Achievements

心筋梗塞や脳梗塞などの動脈硬化性疾患は本邦における死亡原因の上位を占める。本研究では心筋梗塞のリスク遺伝子として同定されたアポA-Vに着目し、新規の動物モデルとしてアポA-V欠損ウサギ作製した。アポA-Vの欠損により中性脂肪に富むリポ蛋白の代謝に異常をきたすことを明らかにした。本研究で樹立したアポA-V欠損ウサギの応用により、脂質異常症や動脈硬化性疾患の新たな予防・治療法の開発が期待される。
