Outline of Research at the Start

Outline of Final Research Achievements
In rats with monocrotaline (MCT)-induced pulmonary hypertension, enhanced phosphorylation and increased sensitivity of pulmonary arterial PKD1 were observed. Inhibition of PKD resulted in a reduction in pulmonary arterial pressure and an increase in cardiac output. Findings from isolated pulmonary vessels and pulmonary artery smooth muscle cells suggested that PKD contributes to pulmonary vasoconstriction via phosphorylation of MYPT1 and MLC. Although siRNA was functional at the cellular level, optimal conditions for in vivo delivery were not established in this study. Nevertheless, our results suggest that pharmacological inhibition of PKD may be a promising therapeutic approach for pulmonary hypertension.

Academic Significance and Societal Importance of the Research Achievements
