脳動脈瘤の形成にエストロゲン(女性ホルモン)が大きく関わっていることが知られている。エストロゲンはエストロゲン受容体βに作用して炎症を抑え脳血管に対して保護的に作用することが考えらている。さらに大豆イソフラボンは腸内細菌でエストロゲン類似物質に代謝されることが報告されている。以上より大豆が腸内細菌によりエストロゲンに代謝され脳動脈瘤の形成を抑制するかを3年間で検証を行う。

Outline of Final Research Achievements |
Postmenopausal women have a higher incidence of intracranial aneurysmal rupture than men of the same age. Equol, a phytoestrogen produced by the gut microbiota from dietary daidzein, is known to preferentially bind to estrogen receptor-beta. The aim of study is to verify equol supplement reduces intracranial aneurysm formation in ovariectomized female mice via gut microbiota. In our experiment, we showed that dietary phytoestrogen significantly reduced intracranial aneurysm formation in ovariectomized female mice. Dietary phytoestrogen with vancomycin had high incidence of aneurysm formation. Equol produced by the gut microbiota from dietary phytoestrogen may play important role in aneurysm formation.

Academic Significance and Societal Importance of the Research Achievements |
腸内細菌を介して生成された女性ホルモン類似物質は脳動脈瘤形成を抑制した結果から、疫学的に閉経前後の女性に多く発症する脳動脈瘤破裂によるくも膜下出血に関して、女性ホルモンが重要な役割を担っていることが本研究を通して示すことができた。脳動脈瘤形成・破裂の高リスクである閉経前後女性への食事療法および栄養学における予防医学観点から重要であり、新しい治療につながることが期待される。
