| Outline of Final Research Achievements |
This study aimed to elucidate the mechanism by which immune cells, specifically macrophages, process urinary crystals non-invasively, thereby establishing a foundation for novel therapeutic strategies. We developed a new in vitro evaluation system using human iPSC-derived macrophages to visualize and quantitatively assess their phagocytic activity against calcium oxalate crystals, clarifying the relationship between macrophage polarization (M1/M2) and phagocytic efficiency. Additionally, we demonstrated that osteopontin binds to the crystal surface and modulates macrophage recognition and response, revealing part of the molecular mechanism underlying crystal clearance. These findings contribute to the development of an immune cell based disease-modifying therapy for urinary stone disease.

| Academic Significance and Societal Importance of the Research Achievements |
