Outline of Research at the Start

胎盤は、母親から供給される栄養素を胎児に渡し、胎児から排出される老廃物を母親に返す役割を担っています。従って、胎盤が正常に発育しなければ、胎児の成長や妊娠の維持に支障をきたします。私達は、胎盤の細胞(栄養膜細胞)のモデル細胞株を用いて、栄養膜細胞の発育の前後の状態を模した細胞をそれぞれ試験管内で増やし、両者を比較することで、栄養膜細胞の発育に必要な因子を見つけ、その因子の働く仕組みを明らかにしようとしています。

Outline of Final Research Achievements

During human placental trophoblast differentiation, the aromatase gene expression is significantly upregulated. To study the transcription mechanism of the aromatase gene in trophoblast cells, we used undifferentiated trophoblast model cell line (JEG-3). We found that a universal methyltransferase inhibitor treatment induces aromatase gene expression and changes chromatin structure in JEG-3 cells.

Academic Significance and Societal Importance of the Research Achievements

胎盤栄養膜細胞のモデル細胞を用いて、アロマターゼ(エストロゲン合成酵素)の遺伝子発現がエピジェネティックに制御される機構の一端を明らかにした。
