Outline of Research at the Start

実験的に血管新生と炎症細胞を誘導したハイリスク角膜移植系を用い、そこで出現する制御性T細胞に対してマルチオミクス解析と機械学習との新しい連携により制御性T細胞の可塑性・不均一性に関わる分子プロファイルの挙動全貌の解明に挑む。この成果により、制御性T細胞の免疫抑制能を低下することなく、移植臓器へ安定的に誘導する免疫誘導経路の確立を目指す。

Outline of Annual Research Achievements

角膜移植は世界で年間60,000件以上行われる最も多い臓器移植であるが、感染症や再移植などで血管新生や炎症が惹起された高リスク角膜に対する移植では40-90%の拒絶反応を伴う。制御性T細胞の挙動が拒絶反応克服の手がかりであるが、炎症などの外部環境の変化により転写因子であるFoxp3が発現を失って免疫抑制能が低下する現象「制御性T細胞の可塑性」に申請者は注目した。ただし、制御性T細胞の不均一な特性がその有効活用を阻んでいる。
そこで本研究は、実験的に血管新生と炎症細胞を誘導した高リスク角膜移植系を用い、そこで出現する制御性T細胞に対してマルチオミクス解析と機械学習との新しい連携により制御性T細胞の可塑性・不均一性に関わる分子プロファイルの挙動全貌の解明に挑む。この成果により、制御性T細胞の免疫抑制能を低下することなく、移植臓器へ安定的に誘導する免疫誘導経路の確立を目指す。
該当年度は、シングルセルRNAシークエンスのT細胞の含まれるクラスターにおける発現変動遺伝子のパスウェイ解析を実施した。IL-2やTGF-βに関する遺伝子パスウェイの関連が明らかになった。また、角膜移植後の角膜移植片を採取し、RNAシークエンスを実施した。高リスク角膜移植では、409遺伝子の増加と150遺伝子の発現の減少が明らかになった。

Reason

該当年度は角膜移植後の角膜移植片サンプルを用いたシングルセルRNAseqならびにRNAシークエンスを実施し、その解析を行った。COVID-19感染拡大によるシークエンス結果の受託の遅延を認めたが、概ね進捗は良好である。

Strategy for Future Research Activity

マウス高リスク角膜移植後の角膜移植片サンプルを用いたシングルセルRNAseqやRNAシークエンスにおける遺伝子発現パターンを機械学習により同定し、免疫細胞の可塑性に関わる遺伝子群や分子経路の特定を行う。
