Outline of Research at the Start

炎症性腸疾患は寛解と再燃をくり返す難病である。患者数は急増し、すでに20万人を超えている。薬剤などにより寛解が得られても、完治させる内科的治療法は存在しない。
我々は、免疫系臓器や腸における支持細胞に脂質が与える影響について検討してきた。すでに、n-3系脂肪酸に親和性の高いFABP7が、リンパ節の支持細胞や各種臓器の免疫細胞を介して炎症を制御しているという結果を得て、報告を行ってきた。今回、「胎児期からの脂肪酸摂取バランスが、炎症の核となる支持細胞の膜の構成やサイトカイン産生を変化させ、炎症細胞の制御を通して寛解や再燃に関与する」という仮説を立て、臨床への応用を図るための基礎研究を試みる。

Outline of Annual Research Achievements

胎児期からの脂肪酸摂取バランスが炎症性腸疾患の寛解と再燃に与える影響を検討するため、我々は多価不飽和脂肪酸のキャリアタンパク質に注目した。このキャリアタンパク質は活性化したB細胞の一部の細胞群に発現しており、炎症制御に関与していると考えている。このキャリアタンパク質を欠損したマウスを用いて、その欠損が炎症性腸疾患に与える影響を検討し、さらに質量分析イメージングにより、その基盤となる分子群の局在変動を解析した。以下、前年までに得られた結果も含めてまとめたものを示す。
方法:(1)KOマウスとWTマウスについて、正常状態の体重や腸を比較した。(2)常法通り、デキストラン硫酸ナトリウム塩(DSS)の自由飲水により腸炎を惹起し、KOマウスとWTマウスの炎症の状態を比較した。(3)質量イメージング解析により、正常状態および炎症部位の大腸のリンパ濾胞に集積する分子群を探索・同定し、その局在をイメージングした。
結果と考察:(1)KOマウスは、WTマウスよりやや体重が多かったが、腸の長さや組織学的な所見には差が見られなかった。(2)DSSを7日間飲水させたところ、KOマウスの腸炎の程度が有意に高くなった。(3)分子局在について比較し、正常状態および炎症部位の大腸のリンパ濾胞に集積する分子の質量イメージング像を得た。この分子は炎症状態のKOマウスのリンパ濾胞にも集積していたが、正常状態より局在量が低下していた。この分子が炎症の惹起や悪化に関与している可能性が高いと考えられた。これらの結果をまとめ、第129回解剖学会全国学術集会で発表した。

Reason

Covid-19の影響で学外との共同研究が進まず計画を大幅に変更したが、それがおおむね順調に進むようになった。

Strategy for Future Research Activity

概要の部分に記した通り、分子局在について比較し、正常状態および炎症部位の大腸のリンパ濾胞に集積する分子の質量イメージング像を得た。この分子は炎症状態のKOマウスのリンパ濾胞にも集積していたが、正常状態より局在量が低下していた。この分子が炎症の惹起や寛解に関与している可能性が高いと考えている。この分子が脂肪酸であると仮定して解析したところ、分子量からいくつかの候補が挙がってきた。候補について標品を準備し、分子の同定に向けた比較を行う予定にしている。
濾胞の細胞については、集積している細胞種の同定を行っている。さらに、上記の解析で同定した分子との反応を解析する方法を検討中でである。
