Outline of Research at the Start

先天性血栓性素因として、アンチトロンビン、プロテインC(PC)、プロテインS(PS)の欠損症がある。先天性血栓性素因のスクリーニング検査として、これらの抗原量および活性の測定が重要である。活性化PC(APC)は、PSを補酵素として、活性化第V因子(FVa)および活性化第Ⅷ因子を不活化し、凝固反応を阻止する。本研究では、現在行われている間接的な活性測定法ではなく、PSおよびPCによるFVaの不活化を直接定量化する測定系の構築を目的とする。本測定法が実現できれば、従来法に比べてより正確にPSおよびPCの機能異常を検出することが可能性になり、血栓性素因の診断に有用であると考えられる。

Outline of Final Research Achievements

We generated recombinant coagulation factor V (FV) mutant as a substrate for measuring protein S (PS) and protein C (PC) activities and established stable cell lines expressing the FV substrate. We established new PS activity assay and PC activity assay using the FV substrate. We measured samples prepared by adjusting the mixing ratio of normal and deficient plasma and successfully detected their activities.

Academic Significance and Societal Importance of the Research Achievements

先天性血栓性素因として、アンチトロンビン、PC、PSの欠損症があり、国の指定難病である。先天性血栓性素因のスクリーニング検査として、これらの抗原量および活性の測定は重要である。現在行われているPSおよびPC活性測定法は、凝固時間法もしくはトロンビンやAPCで分解される合成基質を用いた間接的な測定法であり、本研究で構築した方法は、PSおよびPCの働きを直接定量化することで、より正確に測定できる可能性があり、欠損症の診断に有効である可能性がある。
