Outline of Research at the Start

ヒト循環血中の低分子量ペプチドの構造決定は困難の極みである。我々は独自に開発した血中超微量ペプチド高効率抽出・精製法と超高感度・高精度質量分析を組み合わせた高感度ペプチドミクス技術を完成させ、ヒト血中ネイティブペプチドを構造決定する技術を開発した。これら網羅的に同定したペプチド配列から健常人および各種疾患との比較により、疾患診断や発症・進展予測、治療効果判定のための臨床バイオマーカーとして有用な配列を探索し、臨床的に有用な因子を選択して開発研究を行う。全ての翻訳後修飾情報やアミノ酸置換情報を含めた循環血中ネイティブペプチド同定情報を国際的データーベースとして構築しその始動に貢献する。

Outline of Final Research Achievements

We have succeeded in developing a novel strategy to comprehensively identify low-molecular-weight native peptides using mass spectrometry. Peptides sequences were chemically synthesized to be used for identifying novel bioactive peptides, resulting in identification of two NF-κB-inducers and one anorectic peptide. While analyzing native peptides in human plasma using peptidomics approach, we found potential disease biomarker proteins; oxidative stress markers, a potential cardiovascular disease predictor, and a potential body fluid indicator. This study illustrates a new approach for discovering unknown bioactive peptides and disease biomarkers in plasma via the generation of peptide libraries using a novel peptidomic strategy.

Academic Significance and Societal Importance of the Research Achievements

従来、ヒト血中に存在する超微量ネイティブペプチドの配列を正確に決定することは不可能とされていたが、始めてその網羅的同定に成功し、新たなヒト血漿ネイティブペプチドデータベースを作成して公開した。同定したペプチドの中から3つの強力な生理活性を有する新規生理活性因子を明らかにし、これらが血中に確実に存在するペプチドホルモンであることを示した。さらに、研究の過程で疾患バイオマーカーとなりうる候補因子を3つ同定することができた。これまで新規生理活性因子やバイオマーカーの探索は困難をきわめていたが、「血漿ペプチドーム」はこうした創薬シーズ・マーカー探索研究を加速させる貢献が期待される。
