Outline of Research at the Start

安全な光を用いた分光計測により、体内の生理的変化を計測できる。また近赤外光は、可視光に比べ生体の深部まで到達する。しかし、生体組織における強い光散乱で大きく広がってしまい、体内局所の分光は極めて難しい。
これに対し、位相共役光という特殊な光を生成すれば、あたかも時間を反転したように、光をもと来た経路とは逆に伝搬させることができる。これにより、生体組織で広がった光を逆伝搬させ集束させることも可能になる。
本研究はこの原理を利用し、生体内部局所領域の分光を実現する新たな計測技術を開発しようとするものである。理論解析、シミュレーションにより提案手法を具体化させ、実験によりその実用性の検証をめざす。

Outline of Annual Research Achievements

本研究は、位相共役波による時間反転という新原理を散乱分光の分野に新たに導入することにより、これまで実現が困難であった強い散乱体内部における局所領域分光計測技術の開発を目的とする。2023年度の研究実績は次のとおりである。
1.これまでの結果をもとに、計測方法、計測条件、計測範囲等につき理論解析を進め、いくつかの問題点を抽出した。具体的には、生体のような濃厚系散乱体内部の光伝搬につき、次のように解析を行った。(1)均一散乱体に対しては、光エネルギーの輸送方程式を基本とし、拡散近似により拡散方程式を介して解析解を得た。これにより、具体的な計測システムにおける入射光量、変調周波数、検出感度、計測系のSN比、計測可能深さなどに関し、実現可能性の見通しが得られた。 (2)血管や筋肉層など不均一な対象に対しては、モンテカルロ法に基づくシミュレーションを適用した。二種の異なるアルゴリズムを適用し、結果の一致によりシミュレーションの妥当性を明らかにした。
2.独自に考案したイノベーション、つまり超音波変調周波数を時間掃引するチャープ方式の実用化に向け、検討を深めた。具体的には、 (1) 実用対象について原理解析を行い、チャープ方式を実用レベルでシミュレーションできるようにした。その結果、(2)生体組織のような濃厚系散乱体内部において、光計測領域を局所化する性能が実用レベルに向上することが明らかになった。
3.体内血管のモデルを構築し、イメージング実験を行った。散乱抑制原理を適用することにより、空間分解能が有意に向上することを確認した。これに伴い、提案手法の利点ならびに問題点が明らかになった。
4.これらの成果を国際会議や国内学会で発表した。2023年度は6回の国際会議においてのべ7件の研究発表(Plenary1件、Keynote1件、Invited3件、Top download表彰1件)を行った。

2: Research has progressed on the whole more than it was originally planned.

Reason

体内局所領域の選択的計測やイメージングでは、その空間分解能の優劣が、提案手法成否の鍵となる。従来の方法では、光に対する超音波変調の周波数が一定であり、空間分解能には限界があった。これに対し本研究では、超音波周波数を周期的に掃引したチャープ超音波を用いることを、新たに考案した。
これまでの結果を受け、2023年度は提案手法実用化に向けての具体化を目指した。理論解析を進めるとともに、シミュレーション手法を確立した。また、生体内部血管構造のモデルを作成し、イメージングのシミュレーションを繰り返した。その結果、新たに考案した方法の特長がさらに明らかになるとともに、いくつかの問題点が明確となり、それらの克服を図った。これにより、今後本研究が概ね順調に進展すると考えられる。

Strategy for Future Research Activity

上記のように、これまでの研究は概ね順調に進展している。これらの結果より、位相情報だけではなく強度情報も正しく再現した位相共役波を用いる基本方針は維持する。また、本研究で新たに明らかになった、超音波周波数を掃引する手法をさらに深化発展させる。これらにより、当初の計画を超えた性能が期待される。
今後の実験的検証については、研究代表者の中国大学への転任に伴い、実験設備の輸送に時間を要している。国際情勢の影響により、国際輸送システムや税関検査やが大きく遅滞しているためである。実験設備の一部はすでに中国入り(香港港内)し、現在税関検査待ちの状態である。今後これ等の問題を解決し、極力早期に研究の進展を図る予定である。
