Outline of Research at the Start

阻害剤・機能性分子開発では、主にライブラリー等から取得する必要があり、目的とする結合部位や機能を満たす分子取得には膨大な労力と時間がかかる。従って、標的情報に合わせた精密な分子設計システムは次世代技術として待望されている。そこで、本研究では、標的情報に合わせた化合物とアミノ酸が一体化した融合機能性分子設計システム構築を行う。

Outline of Annual Research Achievements

阻害剤・機能性分子の開発研究では、主にライブラリー等から実験的に取得する必要があり、目的とする結合・相互作用部位や機能を満たす分子取得には膨大な労力と時間がかかる。従って、標的情報に合わせた精密な分子設計システムは次世代技術として待望されている。そこで、本研究では、標的情報に合わせた精密な分子設計システム構築を行う。エンベロープ(細胞由来の脂質二重膜)に覆われた全てのウイルスは膜融合蛋白質を保持しており、膜融合蛋白質はその性質および構造上、必ず疎水性ポケットを持つ。またこのポケットは機能上必須の役割を担う。本研究では、このポケットを標的として、特異的治療法が存在ない高病原性ウイルス感染症を対象に、化合物とアミノ酸が一体化した融合機能性分子を用いて化合物部分の構造とアミノ酸配列を標的蛋白質の構造情報に応じて最適化することで、ウイルス特異的な融合機能性分子を精密にデザインするシステムを構築することを目的に研究を行った。
本年度は、昨年度に標的となるウイルス膜融合蛋白質の立体構造情報に基づき、改良デザインされたペプチド（Chemically-modified peptide: CM-Peptide）の作用機序を理解するため標的蛋白質との構造解析を実施した。また、昨年度とは異なるアプローチとして、CMペプチドを構造情報に基づき適切な距離のリンカーでつないで多価化することによるデザインと合成、感染阻害脳の評価解析を実施した。さらに、抗体による中和機構をペプチドデザインに活かすために抗原―抗体認識の構造生物学的解析も行った。
