Outline of Research at the Start

様々な生命現象を支えるため膜タンパク質の膜への配向は厳密に制御されている。ロドプシンも必ずC末端を細胞質側に向けていることが知られていたが、我々は2018年、膜トポロジーが反転したヘリオロドプシンを発見した。ヘリオロドプシンは正電荷を内側に持っているが、膜反転が静電相互作用だけで説明できるのかどうか不明である。本研究では、アミノ酸変異を駆使して、ロドプシンが膜トポロジーを決める要因を明らかにするとともに、ロドプシンの膜トポロジーを自在に制御することに挑戦する。

Outline of Annual Research Achievements

様々な生命現象を支えるため膜タンパク質の膜への配向は厳密に制御されている。光応答性7回膜貫通型タンパク質であるロドプシンも必ずC末端を細胞質側に向けていることが知られていたが、我々は2018年、膜トポロジーが反転したヘリオロドプシンを発見した。ヘリオロドプシンは構造的にも正電荷を内側に持っているが、膜反転の理由はそれだけによるものか、シグナル配列や第一ヘリックスを含むN末端の性質によるものか不明である。そこで本研究においては、アミノ酸変異を駆使して、ロドプシンが膜トポロジーを決める要因を明らかにするとともに、ロドプシンの膜トポロジーを自在に制御することを目指した。
研究を進める中、同様の研究内容を含む特別推進研究「光遺伝学を支えるロドプシンの作動メカニズムの解明」を2021年度途中から開始したため、萌芽レベルでの研究だけに留まらず、さらに発展した成果が期待される。2021年度、これまでに以下の成果が得られた。
ヘリオロドプシンの精製タンパク質に対して、低温赤外分光や時間分解ラマン分光を行い、光活性化の構造ダイナミクスを明らかにした。また我々は、系統樹においてヘリオロドプシンとC末端を細胞質側に向けた微生物ロドプシンの中間的な位置に存在するシゾロドプシンを見出し、これが内向きプロトンポンプの機能を持つことを明らかにしている。C末端を細胞質側に向けたシゾロドプシンについて、東大との共同研究により立体構造を明らかにした。これら以外にも、C末端を細胞質側に向けた微生物ロドプシン、具体的には新しいチャネルロドプシン、光駆動ナトリウムポンプ、TATロドプシンなどのメカニズム解析を論文発表することができた。これらは膜配向の制御とも大きく関わる成果である。
