Outline of Research at the Start

薬剤耐性菌への対策が急務となっている現在、従来型抗生物質に代わる手段として低温大気圧プラズマが注目されている。本課題では、低温大気圧プラズマの細菌不活化効果の作用機序を解明することを目的とし、海外研究機関において国際標準プラズマデバイスを用いたバイオフィルム状細菌群不活性化実験を行う。多くのプラズマデバイス作動変数を調整することで細菌群に対して非特異的な効果を与えつつ、細菌細胞内のDNA・蛋白質の分子生物学的変化および細菌細胞群の細胞生物学的変化（非耐性菌あるいは耐性菌の死滅や出現の挙動）を解明する。また、細胞内生体反応および細胞群挙動シミュレーションを相補的に用い実験を支援する。

Outline of Annual Research Achievements

抗微生物薬剤に対する耐性を身につけた細菌への対策が世界的な急務となっている現在、従来型抗生物質に代わる手段として低温大気圧プラズマ放電現象が注目されている。本研究は、低温大気圧プラズマの細菌不活化（死滅・感染性を失わせる）効果の作用機序を解明することを目的とした国際共同研究を行う。特に、低温大気圧プラズマの様々な作用変数を制御することで細菌群に対して非特異的な効果を与えつつ、細菌細胞内のDNA・蛋白質の分子生物学的変化および細菌細胞群の細胞生物学的変化（非耐性菌あるいは耐性菌の死滅や出現の挙動）についての定量的な知見を得る。ここでは、特にバイオフィルム状細菌群不活性化実験および細胞群挙動シミュレーションを相補的に用いた研究推進を行う。
令和4年度は、プラズマ生成デバイスからバイオフィルム標的に至る、活性化学種の軌道を明らかにする統合的モデリングの開発に取り組み、低温プラズマと生体ターゲットを繋ぐ知見を得た。
これらの成果に基づき、原著学術論文 1編、国際会議招待講演 2件、国内会議招待講演 1件をはじめ　11件の国際会発表および4件の国内会議発表を行った。

Reason

当初計画以上に数理モデリング研究が進展しており、原著学術論文出版ならびに国内外あわせ3件の招待講演を行った。また実験研究（国内）も順調に進展している。
2020年初頭からつづく新型コロナウィルス感染症に起因した制限が緩和されつつあることを踏まえ、この3年間で停滞した進捗を取り戻すべく、海外研究機関との共同研究体制を再構築する準備を行っている。

Strategy for Future Research Activity

令和5年度は、基礎的実験研究を国内で行うことを基本方針としつつも、状況が許す限りにおいて海外渡航を行う。特に、以下の項目について、数値シミュレーションと実験とのベンチマークを行うことを目標とする。（１）液相化学反応シミュレーションと複雑ネットワーク解析を組み合わせたモデリング手法を構築する。（２）単一細胞レベルの生理化学反応シミュレーションを進展させる。（３）細胞性免疫の主役となる白血球の動的挙動、すなわち遊走・貪食機能に注目した数理モデリングを開発する。（４）低温プラズマ照射が群体レベルでの細胞挙動に与える影響について解明する。
