Outline of Research at the Start

薬剤耐性菌への対策が急務となっている現在、従来型抗生物質に代わる手段として低温大気圧プラズマが注目されている。本課題では、低温大気圧プラズマの細菌不活化効果の作用機序を解明することを目的とし、海外研究機関において国際標準プラズマデバイスを用いたバイオフィルム状細菌群不活性化実験を行う。多くのプラズマデバイス作動変数を調整することで細菌群に対して非特異的な効果を与えつつ、細菌細胞内のDNA・蛋白質の分子生物学的変化および細菌細胞群の細胞生物学的変化(非耐性菌あるいは耐性菌の死滅や出現の挙動)を解明する。また、細胞内生体反応および細胞群挙動シミュレーションを相補的に用い実験を支援する。

Outline of Annual Research Achievements

抗微生物薬剤に対する耐性を身につけた細菌への対策が世界的な急務となっている現在、従来型抗生物質に代わる手段として低温大気圧プラズマ放電現象が注目されている。特に、低温大気圧プラズマによる細胞死の惹起は極めて重要なテーマであり、ここでは活性酸素窒素種のアポトーシス(細胞の自死スイッチ)誘導効果が注目されている。
本研究は、低温大気圧プラズマの細菌不活化(死滅・感染性を失わせる)効果の作用機序を解明することを目的とした国際共同研究を行う。特に、低温大気圧プラズマの様々な作用変数を制御することで細菌群に対して非特異的な効果を与えつつ、細菌細胞内のDNA・蛋白質の分子生物学的変化および細菌細胞群の細胞生物学的変化(非耐性菌あるいは耐性菌の死滅や出現の挙動)についての定量的な知見を得る。
令和5年度においては、海外研究者と共同で開発したプラズマジェットを対象とし、その放電構造を解明するモデルの開発に取り組んだ。また、プラズマ化学種が液相化学反応に及ぼす影響を解明するモデルの開発を行い、低温プラズマと生体ターゲットを繋ぐ知見を得た。低温プラズマによる細胞死の誘導実験を行い、これに対応する単一細胞内のシグナル伝達モデリングを行った。また、海外連携体制の再構築に取り組み、英国・アイルランドに加え、ポルトガル・スロベニア・セルビアの研究者との共同研究の可能性を探っている。
これらの成果に基づき、原著学術論文 1編を著し、国際会議招待講演 7件を行った。加えて、7件の国際会議発表および 7件の国内会議発表を行った。

3: Progress in research has been slightly delayed.

Reason

当初の計画に従って数理モデリング研究が進展しており、原著学術論文1編出版ならびに7件の国際会議招待講演を行った。また実験研究(国内)も順調に進展している。これの成果を踏まえ、2024年度に若手研究者の海外渡航(国際会議における成果公表)を予定している。
一方、2020年初頭からつづく新型コロナウィルス感染症に起因した制限が緩和されつつとはいえ、この3年間で停滞した国際連携活動の進捗を取り戻すことは容易ではない。本課題の期間延長も念頭に置き、海外研究機関との共同研究体制を再構築しつつある。

Strategy for Future Research Activity

令和6年度は、昨年度に引き続き、低温大気圧プラズマの生成する活性酸素窒素種と細胞内生化学種の相互作用システム、および細胞死誘導システムを記述する包括的な生理化学反応数理モデルを構築し数値シミュレーションを行う。現行の反応系(200生化学種・500反応パスウェイ)をさらに拡張し、低温プラズマ照射の特徴である非特性を記述する。特に、低温プラズマの影響として過酸化水素(H2O2)、OHラジカル、NOラジカルなど多彩な活性酸素窒素種のフラックスを考慮するとともに、個々の細胞環境の違いを反映する細胞内タンパク質および酵素(グルタチオン・カタラーゼ等)、個々の細胞の耐性の違いを反映するDNA損傷率を考慮に入れる。ここでは、遺伝情報を司る核および DNA・エネルギー産生と細胞死を司るミトコンドリア・免疫系制御において重要な役割を果たすリガンド・受容体相互作用などを実装し、RONSレベル・局所電界強度などのプラズマ因子とのカップリングを行うことで、正常細胞とがん細胞に対する低温プラズマ曝露効果の選択性についての知見を得ることを目標とする。また、大気圧プラズマジェットの放電構造および低温プラズマの及ぼす液相化学反応の検討も継続する。
一方、新型コロナウィルス感染症に起因して停滞した国際連携活動の進捗を取り戻すことは容易ではない。本課題の期間延長も念頭に置き、海外連携の幅をひろげるべく、英国Queens University Belfast・アイルランドDublin City Universityに加え、ポルトガルUniversity of Lisbon・スロベニアJozef Stefan Institute・セルビアUniversity of Belgrade・ポーランドLublin Institute of Technology の研究者との共同研究体制を確立する。
