Outline of Research at the Start

本研究は，建材からの飛散，浮遊，呼吸に伴う体内吸収までの一連の経気道曝露メカニズムと，発症に至る健康リスクを定量的に予測評価する数値解析モデル(in silico model)を確立し，アスベスト健康被害を最小化するための工学手法開発のための基盤技術を整備する．

Outline of Annual Research Achievements

本研究は建築解体時に発生するアスベストの飛散と空気中濃度を必要十分な工学精度で予測した上で，アスベストの経気道曝露に伴う短期・長期の健康影響を定量的に評価するための数値気道モデルを開発する．2022年度は研究計画に従い，以下の研究課題に取り組んだ．
(1)これまでに開発してきた鼻腔・口腔から気管支第4分岐までの数値気道モデルを拡張し，高精細CTデータを用いて気管支第8分岐まで再現した新たな気道モデルを作製した上で，気管支第9分岐から16分岐までを人工的なアルゴリズムにて再現した終末細気管支モデルを統合した．結果として，鼻腔・口腔から気管支第16分岐(終末細気管支)までの呼吸器詳細幾何形状を再現した数値気道モデルを作成した．
(2)下気道でのアスベスト曝露解析へ適用するため，肺胞位置でのガス交換モデル(酸素－二酸化炭素交換モデル)を作製し，気管支第16分岐位置での境界条件モデルとして統合することで，鼻腔・口腔から気道を経由して肺胞位置までの全ての呼吸器要素を統合した完全呼吸器モデルを作成した．
(3)鼻腔・口腔から気管支第16分岐(終末細気管支)までの呼吸器詳細幾何形状を再現した数値気道モデルを用いて，1nmから10umの広範な粒径を対象とした気道内Lagrange粒子拡散解析を実施することで，上気道から下気道まで含めた粒子沈着分布，上気道・下気道通過確率を解析し，in vivoならびにin vitro結果と比較することで解析精度を検証した．

Reason

研究計画に従い，下気道でのアスベスト曝露解析を可能とする気道モデル開発まで順調に進展している．新型コロナウイルス感染症の蔓延に伴う海外渡航制限も緩和されたことから，2022年度には豪州RMIT大学の共同研究者と対面での共同研究実施も再開された．

Strategy for Future Research Activity

研究の最終年度である2023年度は，新型コロナウイルス感染症の蔓延に伴う海外渡航制限も緩和されたことから，博士課程学生と共に豪州RMIT大学ならびにADRIに滞在し，国際共同研究を現地で対面にて推進する計画である．
