Outline of Research at the Start

本研究では、線虫感染機構に関わる基礎研究を行う。既に得られているサツマイモネコブセンチュウの誘引物質を元に、我々は、既にセンチュウの受容体候補を得ている。様々な線虫類を用いることで、様々なリガンドー受容体を明らかにし、センチュウの宿主選択における進化がどのような分子進化により可能になったかを考察する。

Outline of Annual Research Achievements

植物感染性線虫の農業被害は、年間数十兆円と試算されている。本研究では、植物感染性線虫の感染機構において、植物の線虫誘引活性に注目し、植物と線虫の相互作用に関する研究を展開する。我々は、これまでに、サツマイモネコブセンチュウの植物誘引物質を複数単離してきており、受容体候補も得ている。本申請研究では、多くの線虫類のゲノム情報を用いて、誘引物質ー受容体の組合せに関する情報量を増やし、そのバリエーションについて網羅的に理解することを目的としている。
今年度は、2023年5月から6月にかけ、私に加え、当研究室の古谷准教授、神野博士課程学生、門田研究員(理研)、新屋准教授(明治大)の5名がニース(フランス)のINRAEに赴き、Bruno Favery博士らと研究打合せ、共同研究の実施、シンポジウムの開催などを行った。ここで、センチュウの誘引活性評価の新たな方法を学び、国内でも同様の実験ができる研究環境を整備した。一方、新たに、綿の水抽出物から線虫誘引活性阻害を発見した。当初の研究計画には無かったが、誘引活性物質の単離を開始した。

Reason

2022年度には、相手国側受入教員のBruno Favery博士を熊本に招き、共同研究打合せを行うと共に、国際シンポジウムにより日仏間の研究者コミュニティーの交流を行った。それに続き、2023年度には、研究代表者側から、共同研究者2名(門田研究員(理研)、新屋准教授(明治大))、及び、当研究室の博士課程学生と准教授がフランスに赴き、共同研究の実施、シンポジウムの開催を行う事で、より、研究者コミュニティーの形成が図れた。これまでに、線虫誘引物質を複数単離し、論文発表もできた(Oota et al., 2023; Frontiers in Plant Science)。以上の事から、現在までの進捗状況は概ね順調であると思われる

Strategy for Future Research Activity

2023年度に、新たに、綿の水抽出物から線虫誘引活性阻害を発見した。当初の研究計画には無かったが、誘引活性物質の単離を開始した。2024年度は、この物質の精製を進め、単離・同定を目指す。誘引物質阻害剤は、誘引物質の競合物質である可能性もあることから、この競合活性をも念頭において、精製を進める。
