Outline of Research at the Start

テロメア結合因子として同定されたRif1タンパク質は、代表的な非B型DNA構造であるグアニン4重鎖(G4)に結合する。Rif1は、その他のテロメア因子と相互作用しテロメア長の制御に関与するとともに、染色体腕部にも結合し、ゲノムワイドの複製のタイミングを制御する。Rif1は同時に、DNA二本鎖切断修復、転写も制御する。またRif1は染色体腕部と末端テロメアの相互作用を媒介する可能性が示唆されている。本研究では、腕部におけるRif1-G4/染色体複合体、及びテロメアにおけるRif1を含む核酸-タンパク質複合体の微細構造を解明するとともに、両者の物理上のクロストークを検討する。

Outline of Annual Research Achievements

当初、G4(グアニン四重鎖)DNAと全長Rif1オリゴマーとの複合体の微細形態をクライオ電子顕微鏡による単粒子解析で決定することを目指し、マウスRif1(2,418アミノ酸)及び分裂酵母Rif1(1,400アミノ酸)の大量精製に着手した。しかし、両者とも全長タンパク質として大量精製することは非常に困難であった。
そこで、ヒトRif1の精製を試みた。monoQカラムなどと組み合わせ、比較的濃度の高い標品を得ることができた。マウスRif1についてN末端領域(NTD, 1,151アミノ酸)とC末端領域(CTD, 299アミノ酸)の間にある長い天然変性領域(LID, 968アミノ酸)を欠失させたRif1-NC(NTD+CTD)を293T細胞から大量精製した。また、ヒトRif1-NCの精製を進めた。Huilin Li教授のラボでこれらの電顕観察・単粒子解析を実施している。また、これらの標品を用い高速AFMによりその分子動態を解析した。
N末端92アミノ酸を欠失させた分裂酵母のRif1を精製し、現在Huilin Li教授のラボで電顕観察・単粒子解析を進めている。また。LLPSに関与するアミノ酸を同定したので、その変異体のタンパク調製を行っている。分裂酵母およびヒトRif1のC末端領域ポリペプチドのみでテトラマー(四量体)を形成し、G4にも結合する。これらのポリペプチドを大腸菌で大量精製し、Cryo観察やAFM解析を行った。また、Li博士を訪問し、構造解析についての詳細な打ち合わせを行った。

3: Progress in research has been slightly delayed.

Reason

「研究実績の概要」に記したように、マウスRif1も分裂酵母Rif1も、全長タンパク質として高度に大量精製することは非常に困難であり、これが研究の進捗が遅れた主な要因である。また、内在的な不安定性を有しており、分解産物を除去するのが困難であったことも進歩を遅らせた要因である。また、精製できたタンパク質も電子顕微鏡解析の結果、average画像を作るのが困難であった。おそらく、一部形態に可動性があることが原因であろうと考えた。我々は、C末領域にLLPS形成配列があることを発見し、これが一定の構造を作らないことの原因であろうと考えた。今年度、LLPSを形成しない変異体を、全長およびC末ポリペプチドで作製した。高速AFMの解析から、NTDおよびCTDはダイナミックにその位置関係を変化させていることが明らかになった。さらにG4の存在下では、CTDが分断化されることが明らかとなった。このことはG4結合によりC末が単量体に変換されることを示唆した。
これらのタンパクを用いクライオ電顕による観察を継続するとともに、Rif1C末ポリペプチドを大腸菌から大量精製し、結晶化・構造解析を進めている。

Strategy for Future Research Activity

1) 現在ヒト全長Rif1、Rif1-及び、その変異体を作製しその大量精製を進めている。これらのタンパク質が十分に精製でき次第、電顕観察・単粒子解析に供する。2) また、N末端92アミノ酸を欠失した分裂酵母Rif1、及び、そのC末端のテトラマー形成ドメインについては、Huilin Li 教授のラボで電顕観察・単粒子解析を進め、また、九州大学の神田大輔教授と共同で結晶化、X線構造解析を進める。3) Rif1の分子動態は、その核内染色体構造の制御と大きく関連すると思われる。高速AFMによるRif1のダイナミクスの解析をG4 DNAやリン脂質の存在下で行う。4) IDP領域はリン酸化など種々の修飾を受ける。これらの修飾が分子動態に与える影響を高速AFM観察で明らかにする。5) G4結合が、C末ポリペプチドの多量体形成を阻害し、脂質への結合を促進するメカニズムを解明する。6) IDP領域を欠損するRif1変異株を樹立し、その複製タイミングや染色体高次構造(HiC)に及ぼす影響を解析する。7) Li 教授を訪問し、構造解析の成果の論文化について議論する。
