Outline of Research at the Start

1）異所性骨形成における線溶系酵素の役割の解明、骨化の制御：プラスミノーゲン欠損PlgKOマウス、熱傷モデルマウス、プラスミノーゲンActivatorKOマウスを用いて、異所性骨化における線溶系酵素の役割の解明、骨化の制御を検証する。2）脊椎靭帯骨化と線溶系酵素の関連の解明：PlgKOマウス靱帯骨化モデルを作成し、線溶系酵素、activatorの役割を明らかにする。靱帯骨化症患者の血液サンプルを採取し、靱帯骨化症と線溶系酵素、リン酸代謝の関係を調査する。3）新規骨形製薬の開発：小動物、中動物を用いて化合物Aを局所投与し、投与濃度、投与法を最適化し、前臨床モデルでの検証を目指す。

Outline of Annual Research Achievements

これまでの結果からプラスミン活性が石灰化の出現に重要であることから、PAI-1（plasminogen activator inhibitor-1）はその名の通りtPAを阻害してプラスミン活性を低下させるが、PAI-1inhibitor含有の食餌をこれらのマウスに投与しても石灰化は予防できなかった。つまり石灰化に寄与するplasminogen activatorが主にtPAでなくuPAであることが示唆され、これは昨年行った実験でtPAよりもuPAKOマウスにて石灰化が惹起された結果と一致する。プラスミン活性の主な働きはフィブリン溶解であることから石灰化もフィブリンを介した働きと考え、フィブリノーゲンの変異マウスとプラスミノーゲン欠損マウスを交配し石灰化の出現を観察した。するとフィブリノーゲンの変位の有無で石灰化出現は変わらなかった。つまりプラスミン活性が石灰化に関与するのは線溶系を介してではないことが判明した。
プラスミンはフィブリン溶解だけでなく他の凝固因子や蛋白質を失活させることが知られている。したがってプラスミンの石灰化抑制作用は線溶系ではなくこれらの蛋白質失活による可能性を考え、その対象候補を検索している。
プラスミノーゲン部分KOマウスにおいてカルディオトキシンを下腿三頭筋に注射することで惹起された異所性石灰化を収集し解析した。下腿三頭筋を摘出し、超音波ホモジナイザーで十分に破砕した後、石灰化の比重が大きいことを利用して高濃度スクロース溶液を添加して延伸することで石灰化を組織から単離した。XRDなどで分析すると、石灰化はリン酸カルシウム結晶から成り立っていることが判明した。この結晶構造から多くはハイドロキシアパタイトからなると推定された。

プラスミノンの主役割である線溶系の機能が石灰化とは無関係であることが証明できたことから、プラスミンの他の酵素としての働きに注目すべきであることが分かった。

Strategy for Future Research Activity

プラスミンが作用するフィブリン以外の蛋白質候補から石灰化の機序を解明する。
