Outline of Research at the Start

再生医療は幹細胞を患部に移植をすることで完了するのではなく,移植後の微小環境が重要であり,再生を促進するための微小環境の管理が必須である.本研究では微小環境形成に関与すると考えられるエクソソームに焦点を絞り,骨格筋に対する物理的刺激で放出されるエクソソームがニューロン再生を促進させ脊髄損傷後の運動機能回復に有効であるかを検証するのが目的である.特に再生微小環境の形成には毛細血管ネットワークの退行予防と血管新生促進が不可欠であると考え,骨格筋から放出されエクソソームが脊髄の毛細血管網の構築に関与するかを検証し,再生医療における神経再生の管理のためのリハビリテーションを開発する.

Outline of Annual Research Achievements

本研究では微小環境形成に関与すると考えられるエクソソームに焦点を絞り,骨格筋を刺激することで放出されるエクソソームがマクロファージを制御して,脊髄損傷後の運動機能回復に有効であるかを検証することが目的である. 培養骨格筋細胞へ超音波照射を行い放出されるエクソームを回収して,炎症を惹起したマクロファージに添加した.超音波で誘導されるエクソソームは放出量が増加し,超音波誘導エクソソームはIRG1発現やイタコン酸を増加し,IL-1β,IL-6,TNF-αの発現を抑制した.さらに超音波照射誘導エクソソーム内のmiRNAプロファイル構成をRNAシーケンス解析により非照射と比較するとmiRNAの構成比に相違がなかった.また,炎症を惹起したマクロファージにエクソソームを半減して添加すると,エクソソームの抗炎症効果が観察されなかった.これらの結果から超音波誘導エクソソームの効果はエクソソームの放出量に依存されることが明らかとなった.次にマウスの体外から水中法で超音波照射を実施して,血中のエクソソーム量の増加を確認し,IL-6やTNF-αの発現を抑制することも確認できた.超音波照射は放出されるエクソソームによりマクロファージを制御でき,微小環境を形成できる可能性が示された.また,物理的刺激による他の手段を確立するために低酸素環境による刺激で誘発されるエクソソームに関しても検証を進めている.低酸素環境は骨格筋から放出されるエクソソーム量が増加することやマクロファージへの抗炎症作用も超音波照射と同様に効果があることが明らかになった.尚,国際共同研究先とは電子メールやwebにより打ち合わせを実施した.

3: Progress in research has been slightly delayed.

Reason

骨格筋細胞への超音波照射でエクソソーム放出が促進されることが確認され,このエクソソームがマクロファージを制御し,抗炎症効果があることを確認できた.また,マウスでの検証でもin vitroでの検証と同様の効果が得られた.一方,国際共同研究先に出向いて,モデル動物での検証が必要であるが,国際共同研究先との実験の準備が順調に進まなかったために研究を延長して,準備を進めている.

Strategy for Future Research Activity

骨格筋細胞への超音波照射でエクソソーム放出が促進されることが確認され,このエクソソームがマクロファージを制御し,抗炎症効果があることを確認できた.また,マウスでの検証でもin vitroでの検証と同様の効果が得られた.また,エクソソ-ムの効果を増強させる手段として,他の物理的刺激法も検討中である.今後は骨格筋から放出されるエクソソームや直接受傷部位への超音波照射を利用した脊髄損傷の再生について検証していく計画であり,脊髄損傷モデルの確立も必要不可欠であるため,国際共同研究先と協力して,実施していく計画である.
