Outline of Research at the Start

DNA二重鎖切断誘導後にRNAとDNAからなる特殊な核酸構造R-loopが発生し、その構造が転写共役型相同組換え修復を誘導することで、染色体転座を含むがんゲノム異常の発生を抑制することが判明した。当研究では、「なぜ転写領域において染色体転座が起きるのか」また、もし転写領域に染色体転座が本来的に起きやすいとすれば「転写に関連したクロマチンの構造が遺伝子融合を起こしやすい環境を作っているのか」という問いを立て、転写機構とゲノム異常の発生の関係性を様々な角度から解析することで、がんゲノム異常が発生するメカニズムの解明を目指す。

Outline of Final Research Achievements

In this study, we have investigated why chromosomal translocations occur in transcribed regions and how the genome structures associated with transcription create an environment in which gene fusions are more likely to occur. We found that condensates formed at nucleoli through liquid-liquid phase separation play a critical role in the occurrence of gene fusions at transcriptionally active regions. Thus, we uncover a mechanism by which cancer acquires genome abnormalities.

Academic Significance and Societal Importance of the Research Achievements

当研究によって、細胞内の核小体に形成されるタンパク質の集合体によって、がんを引き起こしうる染色体転座が発生することが判明した。このようなメカニズムは、なぜある特定のタイプのゲノム異常が、特定の種類のがんで起こるのかという問いに対して、一つの答えを与えたと考えられる。
