Research Abstract

運動の重要な要素の一つ「筋収縮」は、インスリンに比肩する強力な糖取り込み促進効果を骨格筋にて発揮するが、その細胞内メカニズムは長く不明のままであった。これまで申請者は、(1)運動/筋収縮はインスリンとは異なる細胞内情報伝達経路で糖取り込みを促進する、(2)AMPキナーゼと呼ばれる分子が糖取り込みの調節に関与している、(3)加えて、筋収縮時の調節にはAMPキナーゼ以外の分子も関与する、(4)その未知の分子はAMPキナーゼと同様にLKB1の下流で働く分子の可能性がある、ことを明らかにしてきた。平成22年度は、LKB1を骨格筋時的にノックアウトした遺伝子組換えマウスを用いて、AMPK-related kinaseファミリー・メンバーの一員であるSucrose nonfermenting AMPK-related kinase(SNARK)が、筋収縮時に糖取り込みを促進させる細胞内情報伝達を媒介する分子であることを明らかにした。以上のことから、骨格筋には少なくとも糖取り込みを促進する細胞内情報伝達経路が3つ存在し(インスリン経路、AMPキナーゼ経路、SNARK経路)、筋収縮および運動による糖取り込みはAMPキナーゼおよびSNARK経路によって調節されていると考えられた。
