Research Abstract

生体関節は、定常歩行運動時にはマイクロレベルを含む弾性流体潤滑により低摩擦・低摩耗を維持し、摩擦増大や表面損傷の発生をもたらす局所的接触が発生しうる薄膜潤滑の場合には、巧みな多種のモードの潤滑機構を有することにより極低摩擦・極低摩耗を実現している。このような潤滑機構は、多モード適応潤滑機構と称されているが、詳細については未解明な点も多い。そこで、本研究では、軟骨表面の複雑な構造の実態を各種の手法で明確化するとともに、極性分子の吸着挙動と水和潤滑効果の相互関係と協調的潤滑機能への寄与を探求した。成果の概要を略述する。
1.水和潤滑に対する生体関節軟骨摩擦面吸着膜形成の影響の解明:除荷・再負荷摩擦試験により軟骨表面水和回復と関節液成分吸着膜の強調作用を明確化できた。2.生体関節軟骨と軟骨細胞の変形・摩擦挙動の実測と数値解析:固液二相体としての関節軟骨の可視化圧縮試験と圧縮及び往復動状態の有限要素解析により、モデル化を可能とするとともに,深さ依存弾性特性・透過の圧密特性・線維スプリング効果などの重要性を把握できた。3.軟骨細胞・滑膜細胞への摩擦刺激・力覚応答試験:圧縮刺激に対する軟骨細胞の応答を明確化した。滑膜組織からの滑膜細胞の単離と培養を可能とし、培養組織の評価を行った。4.軟骨模擬材料および軟骨のマイクロトライボ試験:ポリビニルアルコールハイドロゲル模擬軟骨と関節軟骨の比較試験により、ナノ構造特性の相違の影響を明らかにし、低摩擦化への指針を得た。5.マイクロ血流センサによる軟骨部流動現象の非侵襲計測:標準材料や軟骨におけるシグナルの計測を行い、計測手法の基盤を構築し、酵素処理などの影響を調査した。上述成果について統合的に評価を行うことにより、生体の巧みな多モード自律潤滑機構の未解明部分の複数事象を明らかにできた。
