Research Abstract

【方法】Shati遺伝子またはMock遺伝子を組み込んだアデノ随伴ウイルスベクターを背側線条体もしくは側坐核に注入し、各脳部位特異的にShatiを発現させたマウスを作製した。
【結果】背側線条体にShatiを発現させた(dS-Shati)マウスは、その対照(dS-Mock)マウスと比較して、うつ様症状を評価するtail suspension試験および強制水泳試験における無動時間の有意な延長が観察された。一方、側坐核にShatiを発現させた(NA-Shati)マウスでは、その対照(NA-Mock)マウスと比較して、これらの試験における無動時間に変化は見られなかった。自閉症様行動や社会性行動を評価する新奇物質ケージおよび侵入マウスケージを用いたthree chamber試験において、それぞれの対照マウスでは侵入マウスに対するアプローチ時間の有意な延長を示したが、dS-ShatiおよびNA-Shatiマウスはともに、両ケージに対して同程度のアプローチ時間を示した。新奇環境下での行動量測定による探索行動意欲の検討では、dS-ShatiおよびNA-Shatiマウスは、各対照マウスと比較して、顕著な行動量の変化が見られなかった。また、Y-maze試験における短期作業記憶およびnovel object recognition試験における物体認知能力の検討に関しても、dS-ShatiおよびNA-Shatiマウスは、各対照マウスと比較して有意な変化を示さなかった。
【考察】本研究により、背側線条体におけるShatiは、抑うつに関わる神経機能において重要な役割を果たしていると推察される。また、背側線条体および側坐核の両脳部位におけるShatiは、社会性行動に関わる神経機能に関与していると推察される。今後は、これらの脳部位における神経伝達機能の変化を検討し、上記のような行動変化を導くメカニズムについて検討する予定である。
