Research Abstract

(1)Adiponectin投与にてAPC欠損(Min)マウスの小腸adencmaの個数と大きさは有意に低下した。ヒト、胃癌、大腸癌組織におけるadiponectin受容体(AdipoRI and R2)のmRNA,タンパクの発現量をRT-PCR免疫染色で検討したところ、健常部と比べて共に有意に低下していた。
(2)胃培養癌細胞において、TGF-β添加がAdipoR1とAdipoR2は有意に低下した。
(3)Adiponectinは通常培養液で大腸癌細胞の増殖を抑制したが、グルコース欠乏状態では、細胞の生存を補助する役割を果たし、これはautophagyを介していることが判明した。
以上より、腹腔内脂肪由来のadiponectinは癌の発生、進展を制御する環境因子として重要な役割を有している。一方、TGF-βはadiponectinの受容体を低下させることによって、その抑制作用を回避させる機序を介して癌の進展に促進的な作用を発揮することが推測された。
(4)胃癌術後の内臓脂肪量と皮下脂肪量の変化を臍レベルのCT画像を用いたFATScanにてretrospeotiveに検討すると、迷走神経温存群と切離群との間で、皮下脂肪量の変化には差を認めなかったが、内臓脂肪量は迷走神経温存群で優位に高値に保たれていた。迷走神経は腹腔内脂肪を調節する生理学的機能を有している事が示唆された。
