Research Abstract

本研究の究極の目的は、in vitroでの角膜再生およびその正常構造と機能の維持機構を明確にし、角膜再生の条件を突き止めることを究極の目的とする。そのために、角膜を構成する細胞間の信号系の解析を通じていくつかの課題を明確にすることを申請研究の目的とし、その実施計画として、共培養系を確立した。この共培養はコラーゲンのタイプIで作られた膜を用いることで、細胞間の直接な接触はないが、細胞同士間の情報交換、つまり、各細胞が分泌している因子の隣接している細胞に与える影響を調べるための有効な実験手法である。実際に平成22年度にも、平成21年度につづき、この手法を用い、いくつかの業績を上げている。角膜上皮細胞と角膜実質細胞の共培養で角膜上皮細胞の存在が角膜実質細胞の機能に大きな働きを示した。角膜上皮細胞と共培養した角膜実質細胞では角膜実質細胞の単独細胞培養に比べて、角膜実質細胞内のmatrix metalloproteinases-2(MMP-2)の発現が抑制された。しかし、MMP-1では何にも変化は認められなかった。この様な抑制効果はmRNAレベル、タンパク質レベル両方に見られた。そこで、この様な効果はどういう因子によって調節されるかを培養時のmediumをMulti-Plexにかけて調べた。その結果、角膜上皮細胞から分泌されているinterleukin-1 receptor antagonist(IL-1RA)が関与していることが明らかになった。角膜上皮細胞から分泌されているIL-1RAは角膜実質細胞と共培養することで、その分泌される量は増加していることもわかった。共培養することで、角膜実質細胞から何らかの因子が角膜上皮細胞を刺激し、IL-1RAの発現を上げ、さらに、その刺激され、増加されたIL-1RAにより角膜実質細胞内のMMP-2の発現を調節していることが明らかになった。さらに、この様な発現調節には細胞内のmitogen-activated protein kinase ERKのシグナル経路が関わっていることも突き止めることができた。この様な結果から、角膜上皮細胞は角膜実質細胞の内の重要な機能因子であるMMPの発現を調節することで、角膜の恒常性を維持するために重要な働きをしていることを明らかにした。
