Research Abstract

タイのメジョー大実験養魚池と商用養魚池(ナマズ養魚池とテラピア養魚池)で、microcystin (MC)濃度測定、16SrDNAの全藍藻とMicyorocystis sp.、MC合成遺伝子mcyaDとmcyE(Microcystis特異的)の定量PCRを実施した結果、全項目でテラピア池よりナマズ池が高かった。MC濃度とmcyDコピー数の間にナマズ池で正の相関があったが、テラピア池は明確な相関がなかった。使用したmcyD-primerはMicrocystis以外にPlanktothrixやAnabaenaのmcyDを増幅すること、種毎にMC産生能が異なることから、藍藻種の遷移がMC濃度/mcyD比に影響することもこの原因の一つと推察された。また両池ともにmcy保有藍藻中のMicrocystisは10%以下であり、多くのmcyがPlanktotrixやAnabaena由来であることも示唆された。養魚池から単離のMicrocystisもmcyD非保有株であった。MC-LR/-RR/-YR比もナマズ池とテラピア池では異なり、時間的に変化したことも藍藻種構成の遷移を裏付けた。一方、MC分解遺伝子mlrAコピー数とMC濃度には正の相関があった。霞ヶ浦土浦港に比べ実験養魚池のmlrA/MC濃度比は高く、このことから高いMC分解活性が実験養魚池のMC濃度を低下させ、MC濃度/mcyD比が土浦港より実験養魚池で1/10以下となった原因の一つと推察された。さらに、有毒藍藻(mcyD)発生頻度は全リン濃度に対するロジスティック回帰推定が有効であった。また1μg/L以下のMC-RR,YRとカビ臭物質Geosminと2MIBが養魚から検出され養魚池浄化が必要である為、木炭担体フェンス型生物ろ過装置により藍藻とMCが除去可能なことを示し、藍藻増殖-浄化モデルによって有効な養魚池浄化法を検討した。
