Research Abstract

フラーレン(以下"Flln")各種誘導体の抗酸化活性を比較し各種細胞死への最適な制御法への応用を目指した。(1)Fenton反応、過酸化水素への紫外線B波/A波照射の3種の方法で生成させたHydroxyl radical (・OH)への捕捉能はリポソーム(Lpsm)-FllnがPVP-/_Y-CD-Fllnより大きく経時的保持も優れていてアスコルビン酸(Asc)を凌駕し、Flln不含のLpsm単独では効能が小さかった(DMPO/ESR法)。Fllnの抗酸化容量が大きく、Lpsm中へのFlln分散による・OH捕捉持続力の増強が考えられる。(2)上記(1)の水相に対して油相での抗酸化能はスクワラン(Sqln)-Fllnが優れていてα-TocopherolやAscを凌駕し、Flln不含のSqln単独では効能が小さかった(β-カロテン退色法)。ミクロ環境に依存して最適のFlln誘導体による抗酸化能発揮が考えられる。(3)上記(2)のミクロ環境の他に酸化ストレス剤それ自体の水性/油性の観点から、t-BuOOH, 2, 4-Nonadienal (NDA),過酸化水素の3種によるヒト皮膚角化細胞の細胞死とDNA2本鎖切断はSqln-Fllnで防御され、その大きさはt-BuOOH>NDA>>過酸化水素だった(WST-1法とTUNEL法)。各種酸化ストレス剤とこれ迎撃するFlln誘導体の水溶性/脂溶性に基づく相性が考えられる。(4)酸化ストレス応答性転写因子PPAR-_Yのうち、PPAR-_Y2発現は前駆脂肪細胞OP-9で超高度水酸化Flln(SHHF)で抑制されたが、PPAR-_Y1は差違なく、SHHFよりも水酸化度が小さいFllnは著効でなかった。(5)Sqln-Fllnはヒト皮膚角化細胞HaCaT内部へ半日ほどでほぼ飽和状態まで取込まれ、特に核膜隣接の細胞質に多く分布するが、細胞外排出は数時間で急激に起こった(Flln抗体/免疫染色法)。
