Research Abstract

以下の業績を得た。
1)2型糖尿病モデルラットであるGKラット上腸間膜動脈を用いてAT_1受容体拮抗薬がERK活性を抑制し、内皮由来血管反応を正常化することによりエンドセリン-1誘導血管反応が正常化することを示唆した。また、AT_1受容体拮抗薬が内皮由来化分極因子が誘導する上腸間膜動脈の弛緩反応減弱を是正することを明らかにした。
2)2型糖尿病ラットOLETFラット上腸間膜動脈を用いてAT_1受容体拮抗薬によって是正される内皮依存性血管弛緩反応の減弱のメカニズムにはシクロオキシゲナーゼ(COX)が関与していることを示唆した。
3)2型糖尿病モデルマウスob/obマウス上腸間膜動脈におけるセロトニンが誘導する血管収縮反応の増大には、RhoA/RhoキナーゼやSrcキナーゼの活性増大が関与することを示唆した。
4)疑似高血糖状態を作り出すため、高濃度のグルコース処置によるマウス胸部大動脈の器官培養を行った。すると、gp91 phoxの増大やMn-SODタンパク発現の減少によるスーパーオキサイド産生増加が見られ、その結果PPARγタンパク発現の増加が観察された。
5)STZ誘発糖尿病マウス胸部大動脈を用いて、雌雄差を検討した。その結果、雄の糖尿病マウスでは、アディポネクチン産生が低下し、内皮依存性弛緩反応の減弱が見られた。また、2型糖尿病モデルでは観察されなかったが、STZ誘発糖尿病モデルの雌では、Akt/eNOS経路による弛緩反応の減弱が観察された。
