Diabetes mellitus(DM) is an epidemiological risk factor for human carcinogenesis. In order to clarify the relationship between insulin, the key hormone relevant to DM, and carcinogenesis, we analyzed chemically induced liver and lung carcinogenesis in genetically insulin-deficient, diabetic mice. Relative to euinsulinemia, the insulin deficiency promoted liver tumor development, albeit decreasing lung tumors. Thus, the effects of insulin deficiency were found tissue-specific. The mechanisms of promotion by DM of carcinogenesis should be more complex than generally assumed
