Research Abstract

本研究課題の目的は、第3のガス性メッセンジャーである硫化水素(H_2S)の産生酵素cystathionine-γ-lyase(CSE)の誘導機序とH_2Sの担う細胞機能との関係を、糖尿病に関わりの深い膵B細胞と腎組織を用いて解析し、E_2S産生制御の意義を究明することである。平成23年度の研究では、膵B細胞におけるグルコースによるCa^<2+>依存性CSE発現誘導機序、糖尿病モデルマウス腎組織におけるCSE発現減少とH_2Sによる腎機能保護が明らかになった。阻害薬を用いた薬理学的実験、siRNAを用いた遺伝子ノックダウン実験、ルシフェラーゼレポーターシステムを用いたプロモーターアッセイから、膵B細胞のCSE発現誘導には、Ca^<2+>/calmodulin依存性プロテインキナーゼ(CaMK)II、MAPキナーゼカスケード(MEK-ERK)、ERKによりリン酸化される転写因子Sp1、Elk1が必要であることがわかった。重症糖尿病モデルマウスの腎尿細管では、CSEの発現が減少していること、また、このモデルマウスにH_2SドナーNaHSを投与すると、腎機能が改善する(毛細血管拡張、腎血流量増加)ことがわかった。本年度および昨年度までの成果より、膵B細胞では、糖尿病状態での血糖上昇に応じて、細胞内Ca^<2+>濃度の上昇、プロテインキナーゼ(CaMKII、MEK、ERK)の活性化、さらに、Sp1、Elk1のリン酸化が起こり、CSEの発現が誘導され、その結果産生されたH_2Sが、抗酸化作用を介して膵B細胞の糖毒性を回避する役割を果たすことが明らかになった。腎臓においては、CSEを介したH_2S産生が腎機能の維持に重要であり、また、H_2Sが糖尿病性腎症の治療に有用であることも明らかになった。これらの成果は、私たちの提唱する、H_2Sの内因性ブレーキ(intrinsic brake)説を強く支持すると考えられる。
