Research Abstract

免疫系細胞は造血幹細胞より分化し、骨髄において骨芽細胞や破骨細胞などの硬組織構成細胞と共通の微少環境を形成し、多くのシグナル分子を共有している。破骨細胞分化に必須な共刺激シグナルであるITAM-カルシウム経路に注目し、細胞内カルシウム感知センサーとしてのSOCE(Store-operated Calcium Entry)を詳細に解析することで、造血系細胞のみならず間葉系分化の分子機構についても新しいシグナル伝達経路を提唱できるかどうか検討した。T細胞においてNFATを活性化する因子のスクリーニング系から、SOCEを調節するセンサー分子であるSTIM1/2および細胞膜型チャネルOrail遺伝子クラスターを同定し、C2C12細胞を用いたBMP-2依存性の骨芽細胞分化誘導系、骨髄マクロファージを用いたRANKL依存性の破骨細胞分化誘導系において、STIM1/2・Orail遺伝子の発現を確認した。さらに、RAW264.7からのRANKL依存性の破骨細胞分化が、STIM1/2およびOrailのsiRNAによるノックダウンによって明らかに抑制されることを示した。Orai遺伝子のexon2をpGK-Neoで置換し、3'long armにDTAを挿入したターゲティングベクターを作製し、ES細胞にelectroporationして得られた相同組換え体をblastcyst注入し、独立したキメラマウスを2系統得た。ヘテロマウスの交配によって作出されたホモマウスは新生仔致死性であった。STIM1/STIM2については両遺伝子のdouble floxマウスをCathepsin K CreまたはSox9 Creと交配し、破骨細胞または骨軟骨前駆細胞における表現型の解析をおこなっている。
