Research Abstract |
Chemotherapeutic agent-resistant head and neck cancer cell lines were selected by a dose-escalating exposure to cisplatin or docetaxel. Mitochondrial DNA(mtDNA) from individual drug-resistant cells was extracted and subjected to PCR amplification of full-length mitochondrial genomes to examine mt DNA from single cells. We could detect deleted form of mt DNA in drug-resistant cells. Otherwise, we could not establish mt DNA reproducible amplification from single cells derived by laser micro dissection of fresh frozen head and neck cancer tissues.
